Summary: This study confirms the transcriptional activation function of GlMyb2 during encystation in Giardia lamblia. Through chromatin immunoprecipitation experiments and quantitative real-time PCR, several target genes of GlMyb2 were identified, with a detailed analysis on the promoter region of glcwp1 gene.

Summary: The study identified and localized 46 interactors of polymerized actin in Giardia cells, highlighting the importance of GlActin in multiple cellular processes. Surprisingly, eight of these proteins were found to localize to the ventral disc, an organelle crucial for host attachment in Giardia, suggesting a potential target for new drugs to combat drug resistance in Giardiasis.

Summary: Recent studies have shown that Trichomonas vaginalis can establish persistent infections in the vagina, and this understudied infection can have adverse consequences such as increased risk of HIV-1 infection and cervical cancer. Further research on the spherical form of T. vaginalis called 'pseudocyst' and persistent infections is necessary to improve the diagnosis and treatment of trichomoniasis.

Summary: This study aims to investigate the antigiardial effects of pomegranate polyphenols and demonstrate their inhibitory effects on the growth and adhesion capacity of Giardia lamblia trophozoites. Furthermore, pomegranate polyphenols affect the expression and distribution of α-tubulin in the cytoskeleton of Giardia trophozoites. These findings are significant in the development of potential alternative drugs for giardiasis treatment.

Summary: Giardia lamblia is a major waterborne infection-causing protozoan parasite. The mechanism by which it causes disease is unclear, but it has been found to assemble cholesterol and GM1 glycosphingolipid-enriched lipid rafts that play a role in cyst production. These lipid rafts contain various cellular proteins, including potential virulence factors. Drug treatment experiments showed that these lipid rafts and vesicles play an important role in the spread of giardiasis.

Summary: Attachment to the intestinal epithelium is critical for the lifestyle of Giardia lamblia. The ventrolateral flange is an important structure involved in attachment, and the protein Flangin, which complexes with Giardia actin, plays a role in its formation and function. This study reveals the importance of the unconventional actin cytoskeleton in supporting parasite attachment.

Summary: The genetically related assemblages of the intestinal protozoa parasite Giardia lamblia are morphologically indistinguishable and are often derived from specific hosts. RNA sequencing analysis revealed that the exosomal-like vesicles (ElVs) of each assemblage contained distinct small RNA (sRNA) biotypes. These sRNAs may play a regulatory role in parasite communication and contribute to host-specificity and pathogenesis.

Summary: Ascoviruses are a type of large DNA viruses that infect lepidopteran larvae, and they differ from other plant or animal viruses. Studies have shown that certain host cytoskeletal genes are upregulated after infection, and there is a temporal correlation between their expression and viral vesicle formation.

Summary: Decoding the composition and architecture of the extracellular matrix (ECM) can offer new opportunities to identify diagnostic and therapeutic targets in cancer. Fibronectin and its fibrillary assembly play a crucial role in building the ECM structure and have a significant impact on its characteristics. Studying this unique protein can provide insights into its role as a prognostic and theranostic marker in cancer.

Summary: Actin microfilaments are involved in the regulation of nutrient uptake in root and leaf cells, showing different characteristics in their expression. The co-localization of actin and sucrose transporters in the plasma membrane suggests a potential functional role in sucrose uptake. Pharmacological inhibitors like cytochalasin B and D inhibit sucrose and valine uptake in a concentration-dependent manner.

Summary: The polarisome is a cortical proteinaceous microcompartment that plays a role in organizing the growth of actin filaments and the fusion of secretory vesicles in yeasts and filamentous fungi. The complex between the polarisome subunit Pea2 and the type V Myosin Myo2 anchors Myo2 at the cortex of yeast cells, affecting the formation and localization of the polarisome. Interactions between polarisome-bound Myo2 motors and dynamic actin filaments spatially focus the polarisome and sustain its compact shape, as shown through further experiments and biophysical modeling.

Summary: This study investigated the expression of ATP-binding cassette genes in Giardia trophozoites treated with albendazole or nitazoxanide. The results showed that the ABC-C1 gene was overexpressed in cells treated with both drugs, with higher levels observed with nitazoxanide treatment. This suggests a possible role of ABC-C1 in drug efflux.

Summary: The study showed that terfenadine affects the growth and cell viability of Entamoeba histolytica in a time- and dose-dependent manner, with the most significant inhibitory effects observed with 4 mu M at 48 h. Additionally, terfenadine exhibited low toxicity on Caco-2 cells and significantly inhibited adhesion to Caco-2 cells and erythrophagocytic capacity of the parasite. These findings suggest that terfenadine exerts significant effects on the virulence of Entamoeba histolytica and may be effective in the treatment of amoebiasis.

Summary: The study found that curcumin had no antibacterial activity against Salmonella enterica serovar Typhimurium, even at higher concentrations. Moreover, curcumin caused overgrowth of the bacteria and increased their pathogenicity. It also shortened the lifespan and survival time of Caenorhabditis elegans when they were fed curcumin-treated bacteria. These findings suggest that the indiscriminate use of curcumin should be reconsidered, especially in outbreaks of pathogenic Gram-negative bacteria.

Summary: We conducted a retrospective study on hospitalized patients in Mexico who tested positive for SARS-CoV-2 between June 2020 and January 2022. The study analyzed medical records, demographic data, comorbidities, symptoms, lab findings, and sequencing data. The findings showed differences in patient characteristics, comorbidities, and hospital stays between different waves of the pandemic. The study also revealed the presence of multiple SARS-CoV-2 clades.

Summary: In this study, the cytotoxic effect of terfenadine on Giardia lamblia trophozoites was evaluated. It was found that terfenadine inhibited the growth and cell viability of Giardia trophozoites in a time-dose-dependent manner and caused morphological damage. Importantly, terfenadine showed low toxicity for human intestinal cells, highlighting its potential as a treatment for giardiasis.
