Gender-Related Effects of Sex Steroids on Histamine Release and FcεRI Expression in Rat Peritoneal Mast Cells

Mast cells (MCs) are versatile effector and regulatory cells in various physiologic, immunologic, and pathologic processes. In addition to the well-characterized IgE/Fc epsilon RI-mediated degranulation, a variety of biological substances can induce MCs activation and release of their granule content. Sex steroids, mainly estradiol and progesterone, have been demonstrated to elicit MCs activation. Most published studies have been conducted on MCs lines or freshly isolated peritoneal and bone marrow-derived MC without addressing gender impact on MC response. Our goal was to investigate if the effect of estradiol, progesterone, testosterone, and dihydrotestosterone (DHT) on MCs may differ depending on whether female or male rats are used as MCs donors. Our results demonstrated that effect of sex steroids on MCs histamine release is dose-and gender-dependent and can be direct, synergistic, or inhibitory depending on whether hormones are used alone or to pretreat MCs followed by substance P-stimulation or upon IgE-mediated stimulation. In contrast, sex steroids did not have effect on the MC expression of the IgE high affinity receptor, Fc epsilon RI, no matter female or male rats were used. In conclusion, MCs degranulation is modulated by sex hormones in a gender-selective fashion, with MC from females being more susceptible than MC from males to the effects of sex steroids.

Summary: This study reveals that progesterone may act on Trichinella spiralis eggs by binding to the parasite protein PGRMC2-Ts. The findings have implications in the co-evolution between parasites and hosts, as well as the susceptibility to this infection based on gender. Additionally, these results may contribute to the development of new anti-parasite drugs.

Environmental Pollution to Blame for Depressive Disorder?

Summary: Public concern has emerged about the effects of endocrine-disrupting compounds (EDCs) on neuropsychiatric disorders. Preclinical evidence suggests that exposure to EDCs is associated with the development of major depressive disorder (MDD) and could result in neural degeneration. Studies in Mexico show an increasing incidence of MDD, but no clinical associations have been found between blood levels of EDCs and MDD.

Summary: This study demonstrates for the first time that BPA injection during a specific developmental stage in mice leads to an increase in lung metastasis in adulthood. The analysis of the tumor microenvironment suggests that BPA induces an increase in pro-metastatic cytokines. These findings highlight the potential role of BPA as an inducer of breast cancer metastasis.

Sexual Dimorphism of the Neuroimmunoendocrine Response in the Spleen during a Helminth Infection: A New Role for an Old Player?

Summary: The interaction among the nervous, immune, and endocrine systems is crucial for maintaining homeostasis in vertebrates. This study used the Taenia crassiceps mouse system to investigate the impact of infection on neurotransmitters, cytokines, sex steroids, and sex-steroid receptors in the spleen. The results showed that females had a higher parasite load, while infected males had significantly decreased levels of epinephrine. The expression of IL-2 and IL-4 increased in the spleen of infected mice, but the expression of IL-10 and IFN-gamma decreased. Additionally, differences were observed between the non-infected and infected mice, including increased estradiol levels in infected males and decreased levels in females.

Intratumoral Treatment with 5-Androstene-3β, 17α-Diol Reduces Tumor Size and Lung Metastasis in a Triple-Negative Experimental Model of Breast Cancer

Summary: Breast cancer treatment failure is related to low response rates, high costs, and long-term toxicities. The study found that the androstene-3 beta, 17 alpha-diol (alpha-AED) has potential as a less toxic and effective treatment for triple-negative mammary tumors. In vitro experiments showed that higher doses of alpha-AED have an antiproliferative effect on tumor cells. In vivo experiments demonstrated that intratumoral administration of alpha-AED reduced tumor weight and increased the percentage of natural killer cells in mice tumors. These findings suggest that alpha-AED may be a promising alternative or adjunct therapy for breast cancer.
