Glioblastoma cancer stem cell lines express functional acid sensing ion channels ASIC1a and ASIC3

Acidic microenvironment is commonly observed in tumour tissues, including glioblastoma (GBM), the most aggressive and lethal brain tumour in adults. Acid sensing ion channels (ASICs) are neuronal voltage-insensitive sodium channels, which are sensors of extracellular protons. Here we studied and functionally characterized ASICs in two primary glioblastoma stem cell lines as cell culture models. We detected transcripts of the ACCN2 and ACCN3 genes, coding for ASIC1 and ASIC3, respectively, but not transcripts of ACCN1 (coding for ASIC2). Available microarray data confirmed that ACCN1 is downregulated in glioma. Western blotting confirmed expression of ASIC1 and ASIC3, the most protonsensitive ASICs, in both GBM cell lines. We characterized ASICs functionally using whole-cell patch clamp and detected different types of acid-sensitive currents. Some of these currents had kinetics typical for ASICs and were sensitive to specific toxin inhibitors of ASIC1a or ASIC3, demonstrating that the GBM cell lines express functional ASIC1a and ASIC3 that may enable GBM cells to sensitively detect extracellular pH in a tumour tissue. Microarray data revealed that expression of ACCN2 and ACCN3 is associated with improved survival of patients suffering from gliomas, suggesting that preserved susceptibility to extracellular pH may impair tumour growth.

Summary: DEG/ENaCs interact with neuropeptides via different structural bases and origins, where RFamide neuropeptides activate FaNaCs in invertebrates and modulate ASICs in vertebrates. Dynorphin opioid peptides, on the other hand, stabilize the closed state of ASICs. Despite similar peptide sequences, peptides interacting with DEG/ENaCs from different phyla are evolutionarily unrelated to each other.

Functional analysis in a model sea anemone reveals phylogenetic complexity and a role in cnidocyte discharge of DEG/ENaC ion channels

Summary: Ion channels of the DEG/ENaC family in animals have diverse biological functions. Through phylogenetic analysis, we found a surprising diversity of DEG/ENaCs in Cnidaria. By studying Nematostella vectensis, we discovered an unexpected role of a proton-sensitive DEG/ENaC in cnidocyte discharge. Our study suggests that DEG/ENaCs are versatile channels that have been co-opted for diverse functions in animals since early evolution.

Outcomes and Treatment Approaches for Super-Refractory Status Epilepticus A Systematic Review and Meta-Analysis

Summary: Super-refractory status epilepticus (SRSE) is a condition where status epilepticus (SE) continues or recurs after 24 hours or more of anesthesia, or on the reduction/withdrawal of anesthesia. This systematic review and meta-analysis assessed the clinical characteristics, causes, outcomes, prognostic factors, and treatment approaches for patients with SRSE. The study identified a total of 1200 patients with nonanoxic SRSE and found that long-term treatment was associated with lower mortality but increased risk of moderate to severe disability. Rating: 7/10.

Summary: Elevated impulsivity is a common feature in attention-deficit hyperactivity disorder, bipolar disorder, and juvenile myoclonic epilepsy. This study identified genetic variants associated with impulsivity in juvenile myoclonic epilepsy and found a colocalization with expression quantitative trait loci in the cerebral cortex. Functional characterization in a drosophila model revealed the role of SLCO5A1, a gene coding for an organic anion transporter, in regulating synapse assembly and organization genes. Pathway analysis demonstrated enrichment for presynaptic membrane assembly genes and presynaptic organization genes. The study also showed a genetic correlation between polygenic risk for ADHD and impulsivity scores in juvenile myoclonic epilepsy.

Physiologically relevant acid-sensing ion channel (ASIC) 2a/3 heteromers have a 1:2 stoichiometry

Summary: Acid-sensing ion channels (ASICs) are involved in synaptic transmission and pain sensation. Among the ASIC subunits, ASIC1a and ASIC3 have the highest proton sensitivity, while ASIC2a has lower sensitivity but increases variability by forming heteromers with ASIC1a or ASIC3. In this study, the stoichiometry of the ASIC2a/3 heteromer was investigated. The results showed that only ASIC2a/3 heteromers with a 1:2 stoichiometry had intermediate proton sensitivity, and those with a 2:1 stoichiometry had significantly shifted sensitivity. These findings reveal the physiological relevance and functional differences of ASIC2a/3 heteromers.

Postictal Encephalopathy After Status Epilepticus: Outcome and Risk Factors

Summary: The study aims to investigate the risk factors for the development of postictal encephalopathy and its impact on survival after discharge. The results show that postictal encephalopathy is common after status epilepticus and is associated with poor long-term survival. Seizure characteristics are not independently associated with postictal encephalopathy, while etiology, duration of sedation, age, and premorbid condition are the major risk factors for its development.
