Nanoparticulate vacuolar ATPase blocker exhibits potent host-targeted antiviral activity against feline coronavirus

Feline infectious peritonitis (FIP), caused by a mutated feline coronavirus, is one of the most serious and fatal viral diseases in cats. The disease remains incurable, and there is no effective vaccine available. In light of the pathogenic mechanism of feline coronavirus that relies on endosomal acidification for cytoplasmic entry, a novel vacuolar ATPase blocker, diphyllin, and its nanoformulation are herein investigated for their antiviral activity against the type II feline infectious peritonitis virus (FIPV). Experimental results show that diphyllin dose-dependently inhibits endosomal acidification in fcwf-4 cells, alters the cellular susceptibility to FIPV, and inhibits the downstream virus replication. In addition, diphyllin delivered by polymeric nanoparticles consisting of poly(ethylene glycol)-block-poly(lactide-coglycolide) (PEG-PLGA) further demonstrates an improved safety profile and enhanced inhibitory activity against FIPV. In an in vitro model of antibody-dependent enhancement of FIPV infection, diphyllin nanoparticles showed a prominent antiviral effect against the feline coronavirus. In addition, the diphyllin nanoparticles were well tolerated in mice following high-dose intravenous administration. This study highlights the therapeutic potential of diphyllin and its nanoformulation for the treatment of FIP.

Summary: A variant near the DOCK2 gene on chromosome 5 is associated with severe COVID-19 in patients under 65 years old, with this risk allele being more prevalent in East Asian individuals and rare in Europeans. Decreased expression of DOCK2 is observed in younger patients with severe cases of COVID-19, particularly in non-classical monocytes. Suppressed DOCK2 expression is also found in lung specimens from severe COVID-19 pneumonia patients. Inhibition of DOCK2 function exacerbates the severity of pneumonia in a hamster model of SARS-CoV-2 infection. These findings suggest that DOCK2 plays an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19.

Immunodominant B-Cell Linear Epitope on the VP1 P Domain of a Feline Norovirus Cat Model

Summary: This study analyzed the B-cell linear epitope in the P domain of feline norovirus (FNoV) and found that elevated levels of antibodies against this epitope can improve viral shedding and clinical symptoms in FNoV-infected cats. These findings provide important insights into NoV infections in both humans and cats.

Summary: This study compared the receptor binding affinity and immunogenicity of tag-free and His-tagged SARS-CoV-2 RBD proteins, and found that the tag-free RBD protein had higher affinity for binding and stronger immunogenicity. Additionally, the antibody responses induced by tag-free RBD were more rapid and robust, lasted longer, and demonstrated greater cross-neutralization against the SARS-CoV-2 Delta variant.

Summary: This study demonstrates a facile approach for biomembrane functionalization of planar substrates through intracellular hydrogel polymerization. The resulting cell-gel hybrid exhibits extraordinary stability and retains the structural integrity, membrane fluidity, membrane protein mobility, and topology of living cells. The utility of this hybrid as a tissue engineering feeder substrate is assessed and shown to effectively maintain the stemness of stem cells.

Engineering Cyborg Bacteria Through Intracellular Hydrogelation

Summary: Natural and artificial cells are two common chassis in synthetic biology. Natural cells can perform complex tasks through synthetic genetic constructs, but their autonomous replication often causes safety concerns for biomedical applications. In contrast, artificial cells based on nonreplicating materials provide more defined and controllable functions. The authors created hybrid material-cell entities termed Cyborg Cells, by assembling a synthetic polymer network inside each bacterium, rendering them incapable of dividing. Cyborg Cells preserve essential functions, acquire new abilities to resist stressors, and demonstrate therapeutic potential by invading cancer cells. This work establishes a new paradigm in cellular bioengineering by exploiting intracellular man-made polymers and their interaction with living cells' protein networks.

Summary: The study found that specific-pathogen-free laboratory cats developed cross-reactive antibodies to the receptor binding domain (RBD) of human SARS-CoV-2 virus after mating with feline coronavirus-positive queens. These antibodies also showed cross-reactivity with two serotypes of feline coronavirus and could inhibit feline coronavirus infection.

Suppression of myeloid YAP antagonizes adverse cardiac remodeling during pressure overload stress

Summary: Inflammation is a crucial factor in cardiovascular disease, and Yes-associated protein (YAP) has been found to play a key role in modulating inflammation in ischemic injury. This study investigated the role of YAP in non-ischemic cardiac inflammation. The results showed that YAP activation in myeloid cells contributes to cardiac inflammation and fibrosis during pressure overload stress. Inhibition of YAP may be a potential therapeutic target for non-ischemic heart disease.

Expanding the range of the respiratory infectome in Australian feedlot cattle with and without respiratory disease using metatranscriptomics

Summary: This study used metatranscriptomics to identify the pathogens responsible for bovine respiratory disease, including bovine nidovirus, influenza D, bovine rhinitis A, and bovine coronavirus. Complete or near-complete genomes of bovine rhinitis B, enterovirus E1, bovine viral diarrhea virus, and bovine respiratory syncytial virus were also obtained. A new species of paramyxovirus was identified. This research underscores the importance of studying previously uncultured viral species in cattle respiratory disease.
