Summary: Helicobacter pylori infection induces a strong inflammatory response in gastric mucosa and alters epithelial integrity and function. The activation of NF-κB transcription factors, specifically by the bacterial heptose metabolites, triggers this innate immune response. Our study reveals that the activation of ALPK1-dependent TIFA in H. pylori-infected gastric epithelial cells is followed by TIFA degradation, which is inhibited by proteasomal and lysosomal inhibitors. TRAF2, TRAF6, TAK1, or NEMO are not involved in TIFA degradation, and H. pylori promotes the interaction of TIFA with protein adaptors involved in lysosomal trafficking.

Summary: This study reveals that early-stage infection with various viruses, including influenza A virus, leads to IL-6-independent phosphorylation of STAT3 Y705, which is critical for antiviral immunity. Knockin mice with STAT3Y705F/1 mutation show suppressed antiviral response, lung tissue injury, and poor survival upon IAV infection. The phosphorylation of STAT3 Y705 restrains IAV pathogenesis by repressing excessive IFN production, and blocking the phosphorylation enhances IFN expression and potentiates IAV virulence in mice. Knockout of IFNAR1 or IFNLR1 in STAT3Y705F/1 mice protects them from lung injury and reduces viral load.

Summary: Through studying an experimental colitis mouse model, it was found that the increased systemic exposure of metronidazole in patients with inflammatory bowel diseases might be related to the downregulation of CYP2A5, which is regulated by the inflammatory factor NF-KB. The regulatory effect of colitis on CYP2A5 depends mainly on the disease itself, rather than the inflammatory compound, DSS.

Summary: We investigated the reactivation of quiescent cells through a zebrafish model and observed an increase in mitochondrial membrane potential in the reactivated cells. Genetic and pharmacological perturbations revealed that elevated mitochondrial metabolism and ATP synthesis are crucial for cell reactivation. Further analysis showed that the elevated mitochondrial metabolism leads to increased mitochondrial ROS levels, which induce the expression of Sgk1 in the mitochondria. Sgk1, in turn, promotes S phase entry in human breast cancer cells by coordinating mitochondrial activity with ATP synthesis through phosphorylating F1Fo-ATP synthase.

Summary: This study aimed to explore the pharmacological properties of zingerone (ZO) in attenuating osteoclast-mediated periprosthetic osteolysis and how ZO modulates osteoclas-togenesis. The results showed that ZO has an obvious inhibitory effect on osteoclastogenesis and bone resorption in a dose-dependent manner by mainly suppressing the activation of NF-KB signaling pathways. Furthermore, ZO administration successfully attenuated titanium (Ti) particle-stimulated periprosthetic osteolysis and osteoporosis by regulating osteoclast formation.

Summary: This study aimed to investigate the impact of gastrointestinal digestion/Caco-2 cell transport on the biological activities and toxicity of ethanolic extract of organic propolis from southern Brazil (EEOP1). The results demonstrated that EEOP1 deactivated reactive oxygen species and attenuated the activation of transcription factors, while also reducing the release of pro-inflammatory cytokines. Additionally, the phytochemical profile of EEOP1 changed after digestion, and antimicrobial activity was only observed in the non-digested form.

Summary: This study investigated the epigenetic effects of MUC17 in acquired drug-resistant cells of EGFR-TKIs. In gefitinib/osimertinib-resistant (GR/OR) cells, downregulation of MUC17 was mediated by promoter methylation promoted by the DNMT1/UHRF1 complex, leading to activation of NF-kappa B activity. MUC17 acted as an epigenetic sensor for monitoring resistance to EGFR-TKIs in GR/OR cells.

Summary: Most tumors, including brain tumors, occur sporadically. However, there is a small subset of central nervous system tumors that are associated with Lynch Syndrome. This report presents a rare case of an oligodendroglioma with a PMS2 mutation associated with Lynch Syndrome. The limited literature on germline PMS2 mutations and oligodendrogliomas highlights the importance of genetic testing in neuro-oncology.
