Summary: The epidemiology of Clostridioides difficile infection (CDI) has changed over the past two decades, with the emergence of clinically relevant C. difficile strains causing severe nosocomial outbreaks. This study reports on an outbreak that occurred in a Long-term Care Unit, which was tracked using a Matrix-Assisted Desorption/Ionization Time-of-Flight Mass Spectrometry (MALDI-TOF MS) typing approach (T-MALDI). The study also characterized the toxigenic and antimicrobial susceptibility profiles of the C. difficile isolates. The results showed the presence of three different clusters, with one cluster being considered epidemic and susceptible to vancomycin and metronidazole but resistant to moxifloxacin. This is the first reported outbreak associated with a tcdA+/tcdB-/cdt+ genotypic profile. T-MALDI is shown to be a valid and rapid approach for detecting and tracking outbreaks.

Summary: Glycosylation, a common post-translational modification in mammals, particularly N-glycosylation, plays a crucial role in various processes during the development of the mammalian central nervous system. N-glycosylation changes have been observed in neuropathological conditions such as epilepsy and Alzheimer's disease. In this study, N-glycan chains were extracted from different regions of the mouse brain and analyzed using high-throughput MALDI-TOF/MS combined with Fmoc-OSU derivatization to improve glycan detection sensitivity. The study identified 96 N-glycan moieties, including high mannose, fucosylated, sialylated, and galactosylated N-glycans, with significant differences between regions. The comprehensive mapping of glycans in the brain can provide insights into glycan-mediated mechanisms and potential biomarkers for brain-associated pathologies.

Summary: In this study, we developed a matrix-assisted laser desorption ionization-Fourier transform ion cyclotron resonance mass spectrometry (MALDI-FTICR MS) strategy for the accurate characterization of different types of micro/nanoplastics. By optimizing the sample preparation and detection conditions, we improved the detection signal-to-noise ratios and achieved unambiguous elucidation of polymer constitutions of environmental micro/nanoplastics.

Summary: This study used MALDI-MSI to investigate the spatial distribution of lipids in cottonseeds, revealing heterogeneous distribution patterns of different lipid species within the seeds. The roles of these lipids in metabolic pathways were analyzed, and the relationship between their spatial distribution and functions was studied. The unique distribution patterns of lipid metabolites revealed by MSI may offer new insights into the spatial compartmentation of lipid metabolism in plants and can potentially aid genetic engineering efforts when combined with transcriptomics and proteomics.

Summary: GC x GC-O-TOF-MS and GC-IMS have been increasingly used in complex food flavor analysis, but few studies have applied these techniques to identify the aroma components of tomato pastes. This study comprehensively characterized the aroma profiles of cold and hot break tomato pastes using SAFE-GC-O-MS, SAFE-GC x GC-O-TOF-MS, and HS-GC-IMS for the first time. A total of 274 volatile compounds were identified, which is far more than previously reported.

This method provides important information for identity testing, sequence validation, and impurity analysis.
