Summary: Adipose-derived stem cells (ASCs) are a valuable resource for regenerative medicine and research on fat deposition. However, the isolation procedure of ASCs needs to be standardized and the differences in their proliferation and adipogenic differentiation potential from different fat depots require further understanding. This study compared the efficiency of ASCs isolation methods, with explant culture method showing simplicity and lower cost compared to the complex and costly enzymatic treatment method. ASCs isolated by the explant culture method exhibited better proliferation and adipogenic differentiation potential, although slightly lower than those isolated by the enzymatic treatment method. ASCs isolated from visceral depot demonstrated higher proliferation and adipogenic differentiation potential.

Summary: The investigation of mesenchymal stem cells has become an interesting scientific field in regenerative medicine and organ transplantation. This study focused on establishing an adipogenic differentiation protocol suitable for different mammalian species, and found that using rosiglitazone instead of traditional protocols significantly improves cell viability and adipogenic differentiation capacity. The study also revealed differences between species in terms of fat formation and fat-regulating gene expression. This information is essential for selecting a stem cell specification program and improving clinical targeting of obesity and metabolic disorders in veterinary medicine.

Summary: Adipose-derived mesenchymal stromal cells (ASCs) have the potential to differentiate into various cell types and exhibit spontaneous Ca2+ oscillations. The effects of differentiation treatments on ASCs' Ca2+ dynamics, in terms of oscillation fraction and frequency, were examined in this study. Both adipogenic and osteogenic differentiation resulted in reduced Ca2+ dynamics, with adipogenic differentiation showing a more pronounced effect. Changes in Ca2+ oscillations were observed as early as 7 days in vitro (DIV). Further research is needed to understand the molecular and mechanistic correlates of these changes for the development of interventions in regenerative medicine.

Summary: Cultured meat is an efficient and sustainable technology, and adipose-derived stem cells (ADSCs) show promise for its production. This study found that the proliferation and adipogenic differentiation of ADSCs decreased significantly during serial passage. It also demonstrated that N-Acetylcysteine (NAC) can enhance the proliferation and maintain the differentiation of ADSCs. Additionally, NAC was shown to restore the cell cycle and DNA repair pathway in ADSCs, making it an excellent supplement for large-scale expansion of ADSCs for cultured meat.
