A bulk sub-femtoliter in vitro compartmentalization system using super-fine electrosprays

The extreme miniaturization of biological and chemical assays in aqueous-droplet compartments enables spatiotemporal control for large-scale parallel experimentation and can thus permit new capabilities for digitizing directed molecular evolution methodologies. We report a remarkably facile bulk method to generate mega-scale monodisperse sub-femtoliter aqueous droplets by electrospray, using a prototype head with super-fine inkjet technology. Moreover, the electrostatic inkjet nozzle that injects the aqueous phase when immersed within an immiscible phase (an optimized oil/surfactant mixture) has the advantage of generating cell-like sub-femtoliter compartments for biomolecule encapsulation and successive biological and chemical reactions. Sub-femtoliter droplets of both liquid (water-in-oil, volumes ranging from 0.2 to 6.4 fL) and gel bead (agarose-in-oil, volume ranging from 0.3 to 15.6 fL) compartments with average sizes of 1.3 mu m and 1.5 mu m, respectively, were successfully generated using an inkjet nozzle at a speed of more than 105 droplets per second. We demonstrated the applicability of this system by synthesizing fluorescent proteins using a cell-free expression system inside electrosprayed sub-femtoliter droplets at an accelerated rate, thereby extending the utility of in vitro compartmentalization with improved analytical performance for a top-down artificial cellular system.

Determination of alkali and alkaline earth elements in radioactive waste generated from reprocessing plant by liquid electrode plasma optical emission spectrometry

Summary: A simple and reliable analytical method using LEP-OES was developed for determination of Na, K, Ca, Sr, and Ba in radioactive waste, with investigation on emission lines, interference from co-existing elements, and adjustment of nitric acid concentration. Spike and recovery tests showed negligible sample matrix effect, with data matching with ORIGEN code and inductively coupled plasma optical emission spectrometry within 10% difference, demonstrating successful application to real radioactive wastes.

DOCK11 and DENND2A play pivotal roles in the maintenance of hepatitis B virus in host cells

Summary: Using single-cell RNA sequencing analysis, four genes including DOCK11 and DENND2A were identified to be linked to the maintenance of HBV in liver cells. Knockdown of these genes in primary human hepatocytes infected with HBV resulted in a decrease in both HBV DNA and covalently closed circular DNA. This study reveals a role for DOCK11 and DENND2A in maintaining HBV in a latent state.

Electrical properties and parameter analysis of all-solid-state LaZrO/InO electric double-layer transistors

Summary: All-solid-state electric double-layer (EDL) thin-film transistors (TFTs) with LaZrO gate insulators and sputtered InO channels were studied. The transfer conductance of the TFTs increased with the thickness of the InO layer and reached a maximum at 200 nm. The capacitance of LaZrO indicated the formation of an EDL. The mobility of the TFTs also increased with the InO thickness and was affected by grain boundary and surface scattering.

Facile formation of giant elastin-like polypeptide vesicles as synthetic cells

Summary: Giant peptide vesicles were generated easily and robustly using an emulsion transfer method, able to withstand chemical and physical stresses while hosting cell-free expression reactions. Additionally, another cell-free expressed elastin-like polypeptide was successfully incorporated into the existing membrane of the peptide vesicles.
