Summary: RIP1 plays a crucial role in the metastasis of colorectal cancer by controlling the WNT/beta-catenin signaling pathway. The study shows that WNT3A treatment increases the expression of RIP1 and beta-catenin, and elevated RIP1 expression is associated with carcinogenesis and metastasis. In vitro experiments also demonstrate that RIP1 promotes migration and invasion of cancer cells by enhancing endothelial-mesenchymal transition (EMT) through the WNT3A-RIP1-beta-catenin pathway.

Summary: This study provides a detailed overview of gene expressions when human mesenchymal stem cells are cultured on graphene surfaces derived from either adipose tissue or bone marrow. Results show that both MSC sources achieved osteogenesis on graphene surfaces by 21 days, but exhibited genetically different responses. Additionally, upregulated angiogenic markers suggest graphene could attract ingrowth of blood vessels in vivo.

Summary: This study reveals the impact of normal oxygen levels on the differentiation of bone-derived mesenchymal stem cells. High oxygen concentration promotes chromatin compaction and histone hypo-acetylation, resulting in osteogenic defects. Additionally, decreased activity of citrate carrier leads to the accumulation of acetyl-CoA inside mitochondria. Restoring cytosolic acetyl-CoA levels rescues the osteogenic defects.

Summary: Idiopathic pulmonary fibrosis (IPF) is a progressive lung disease, and pulmonary endothelial cells may generate pulmonary fibroblasts through endothelial mesenchymal transition (EndoMT), leading to pulmonary fibrosis. The tyrosine kinase receptor inhibitor, nintedanib, has been found to ameliorate bleomycin-induced pulmonary fibrosis and suppress EndoMT in vivo and in vitro models. These findings contribute to a better understanding of the mechanism and signaling pathway of EndoMT, and provide new insights for developing more effective drugs for IPF treatment.

Summary: RSPO2 promotes the growth and metastasis of ovarian cancer through the activation of FAK/Src signaling cascades. It prevents the endocytic degradation of LGR4 and promotes LGR4-mediated translocation of Src to the plasma membrane. In addition, it directly binds to integrin beta 3 and enhances the stability of integrins. High expression of RSPO2 is associated with poor prognosis in ovarian tumors.

Summary: EGFL6 was found to stimulate angiogenesis and osteogenic differentiation through the Wnt/beta-catenin signaling pathway in vitro, and promoted CD31(hi)EMCN(hi) type H-positive capillary formation in a rat tibia distraction osteogenesis model. It enhanced the osteogenic differentiation potential of BMSCs and accelerated bone regeneration by stimulating angiogenesis.

Summary: This study investigated the osteogenic properties of phenolated alginate hydrogel containing collagen and nano-hydroxyapatite on human mesenchymal stem cells. The results showed that the combination of alginate/collagen/nano-hydroxyapatite had suitable mechanical features and promoted cell viability, osteoblast differentiation, and new bone formation through involving the Wnt signaling pathway.

Summary: Integrin Adhesion Complexes (IACs) act as links between the cytoskeleton and extracellular environment, participating in cellular motility, tissue morphogenesis, anchorage-dependent growth, and cell survival. Focal Adhesion Kinase (FAK) is a critical organizer of IAC signaling events and a genetic and therapeutic target. This study presents the design and characterization of reversible Bimolecular Complementation sensors to monitor FAK phosphorylation in living cells, providing novel means to quantify IAC signaling.
