Urinary candidate biomarker discovery in a rat unilateral ureteral obstruction model

Urine has the potential to become a better source of biomarkers. Urinary proteins are affected by many factors; therefore, differentiating between the variables associated with any particular pathophysiological condition in clinical samples is challenging. To circumvent these problems, simpler systems, such as animal models, should be used to establish a direct relationship between disease progression and urine changes. In this study, a unilateral ureteral obstruction (UUO) model was used to observe tubular injury and the eventual development of renal fibrosis, as well as to identify differential urinary proteins in this process. Urine samples were collected from the residuary ureter linked to the kidney at 1 and 3 weeks after UUO. Five hundred proteins were identified and quantified by LC-MS/MS, out of which 7 and 19 significantly changed in the UUO 1-and 3-week groups, respectively, compared with the sham-operation group. Validation by western blot showed increased levels of Alpha-actinin-1 and Moesin in the UUO 1-week group, indicating that they may serve as candidate biomarkers of renal tubular injury, and significantly increased levels of Vimentin, Annexin A1 and Clusterin in the UUO3-week group, indicating that they may serve as candidate biomarkers of interstitial fibrosis.

Proteomics applications in biomarker discovery and pathogenesis for abdominal aortic aneurysm

Summary: Proteomics has been used to identify potential biomarkers, understand pathogenic mechanisms, and discover therapeutic targets for AAA. However, validation in multicenter cohorts is needed for promising biomarkers to accelerate their clinical application. The development of mass spectrometry technology, modification-specific proteomics, and multi-omics research will enhance our understanding of AAA pathogenesis and facilitate biomarker discovery and drug development for clinical translation.

Proteomic Profiling of Cryoglobulinemia

Summary: This study aimed to identify and validate candidate protein biomarkers for cryoglobulinemia (CGE) using a TMT-PRM-ELISA workflow. APOA1 and PCSK9 were confirmed to be increased in CGE patients, indicating the involvement of proteins in cholesterol metabolism in the development of CGE. These findings provide valuable insights for pathogenesis research and biomarker discovery in CGE.

Comprehensive analysis of tissue proteomics in patients with papillary thyroid microcarcinoma uncovers the underlying mechanism of lymph node metastasis and its significant sex disparities

Summary: This study investigated the molecular mechanisms underlying lymph node metastasis (LNM) and its sex disparities in papillary thyroid microcarcinoma (PTMC) using a proteomics approach. The results showed that LNM in PTMC was associated with mitochondrial dysfunction and activation of oxidative phosphorylation and PI3K/AKT signaling pathways. The study also found distinct proteomics profiles between male and female PTMC patients with LNM, with elevated levels of extracellular matrix-associated proteins in male patients. Furthermore, certain proteins were identified to be downregulated and correlated with tumor metastasis, while others were upregulated and correlated with sex disparities and poor prognosis. These findings enhance our understanding of the functional networks and signaling pathways related to PTMC with LNM.

This study lays a foundation for early detection of high-risk LUAD patients.

Investigation of the Pathogenic Mechanism of Ciprofloxacin in Aortic Aneurysm and Dissection by an Integrated Proteomics and Network Pharmacology Strategy

Summary: This study investigated the functional mechanism and molecular targets of fluoroquinolones in relation to aortic aneurysm and dissection (AAD) through proteomic and network pharmacology analysis. Differential protein expression was observed in human aortic vascular smooth muscle cells (VSMCs) after ciprofloxacin (CIP) stimulation, highlighting the involvement of metabolism, extracellular matrix homeostasis, mitochondrial damage, focal adhesion, and apoptosis. Four critical target proteins, PARP1, RAC1, IGF1R, and MKI67, were identified through protein-protein interaction analysis. Signaling pathways such as MAPK, focal adhesion, apoptosis, actin cytoskeleton regulation, and PI3K-Akt were found to be significantly enriched. These findings provide novel insights into the pathogenic mechanism of fluoroquinolones in aortic diseases.

Identification of Novel Plasma Biomarkers for Abdominal Aortic Aneurysm by Protein Array Analysis

These biomarkers have the potential to be therapeutic targets for AAA and further investigation is needed.

Plasma Lipidomics Analysis Reveals the Potential Role of Lysophosphatidylcholines in Abdominal Aortic Aneurysm Progression and Formation

Summary: Abdominal aortic aneurysm (AAA) is characterized by irreversible dilation of the infrarenal aorta. This study explored the variation of lipids during AAA development and found that lysophosphatidylcholines (lysoPCs) were specifically decreased in AAA patients and mice. The study also identified weakened positive correlations between lysoPCs and HDL-c in AAA patients, suggesting that HDL-lysoPCs may play a role in AAA pathogenesis. These findings suggest that lysoPCs could be promising biomarkers for AAA development.

Urinary complement proteins in IgA nephropathy progression from a relative quantitative proteomic analysis

Proteomics analysis of carotid body tumor revealed potential mechanisms and molecular differences among Shamblin classifications

Summary: Carotid body tumors (CBTs) are a rare type of paraganglioma that can only be effectively treated through surgical resection. This study aimed to investigate the molecular differences between different CBT types and identify potential therapeutic targets. Through proteomics analysis, the researchers found significant differences in protein profiles among different Shamblin subtypes of CBT. They also identified several signaling pathways that are significantly enriched in CBT. Furthermore, the researchers identified key differentially expressed proteins (DEPs) and validated them using immunohistochemistry. This study provides valuable insights into the pathogenesis of CBT and may contribute to the development of targeted therapies.

Early urinary candidate biomarkers and clinical outcomes of intervention in a rat model of experimental autoimmune encephalomyelitis

Summary: Multiple sclerosis is a chronic autoimmune demyelinating disease of the central nervous system that is difficult to diagnose in early stages. A study analyzed the early urinary proteome changes in a multiple sclerosis rat model and identified potential biomarkers for early detection. Functional analysis showed that these proteins are associated with asparagine degradation, neuroinflammation, and lipid metabolism. Early treatment with methylprednisolone significantly reduced the incidence of encephalomyelitis. Urine may be a good source of biomarkers for early diagnosis of multiple sclerosis.

Nomograms Based on Serum N-glycome for Diagnosis of Papillary Thyroid Microcarcinoma and Prediction of Lymph Node Metastasis

Summary: This study characterized the N-glycome of papillary thyroid microcarcinoma (PTMC), and established nomograms for the diagnosis of PTMC and the prediction of lymph node metastasis (LNM). The study found significant differences in serum N-glycome between PTMC and healthy controls, as well as associations between certain N-glycan traits and PTMC and LNM. Nomograms based on these traits showed good performance in the identification of PTMC and the prediction of LNM.
