The impact of p53 in predicting clinical outcome of breast cancer patients with visceral metastasis

In the study, we analyzed role of p53 in predicting outcome in visceral metastasis breast cancer (VMBC) patients. 97 consecutive VMBC patients were studied. P53 positivity rate was 29.9%. In the p53-negative group, median disease free survival (DFS), and time from primary breast cancer diagnosis to death (OS1), time from metastases to death (OS2) were 25, 42.5, and 13.5 months, respectively. In the p53-positive group, they were 10, 22, and 8 months, respectively. Statistically significant differences in DFS and OS1 were detected between the p53-negative and p53-positive subtypes. However, p53 appears to have no influence on OS2. In Cox regression analysis, p53 expression and TNM stage were predictive factors of DFS. In the multivariate analysis, p53 expression and the duration of DFS correlated with OS1, but not for OS2. Taken together, our data indicate p53 showing predicting role in OS1 for VMBC, but not for OS2.

The probe exhibited outstanding targeting ability both in vitro and in vivo, enabling more complete tumor resection, improved survival rate, and differentiation between benign and malignant breast tissues. Additionally, the design of the probe reduced immune response.

Summary: A novel self-assembled multifunctional nanoprobe (NPs-Bev) is developed to improve the efficacy of fluorescence-image-guided breast-conserving surgery (BCS) and radiation therapy (RT) for breast cancer treatment. The NPs can accurately discriminate between benign and malignant mammary tissue, and induce more reactive oxygen species under X-ray radiation therapy, leading to increased sensitivity and reduced tumor progression in mice. The NPs, composed of clinically safe reagents, provide a solid precision therapy platform for breast cancer treatment.

Clinical applications of deep learning in breast MRI

Summary: Deep learning, a powerful data-driven machine learning technique in AI, has shown remarkable advances in data extraction and analysis for medical imaging, particularly in breast MRI. This review summarizes the current applications of DL models in breast MRI, including advanced learning approaches and methods for data normalization and breast and lesion segmentation. DL-based breast MRI models have been proven useful in various clinical aspects, such as breast cancer diagnosis, molecular and histopathological type classification, prediction of chemotherapy response, and lymph node metastasis prediction. Further improvements in data acquisition and preprocessing, exploration of additional DL techniques, and wider clinical applications are needed for future studies.

Notch1 promotes resistance to cisplatin by up-regulating Ecto-5′-nucleotidase (CD73) in triple-negative breast cancer cells

Summary: This study explored the molecular mechanisms of chemoresistance in triple-negative breast cancer (TNBC) and found that Notch1 and CD73 were associated with poor clinical outcome in cisplatin-treated patients. Further experiments revealed that Notch1 regulated CD73 expression by directly binding to the CD73 promoter. These findings suggest that CD73 is a direct downstream target of Notch1 in mediating cisplatin resistance in TNBC.

In vivo visualization of fluorescence reflecting CDK4 activity in a breast cancer mouse model

Summary: A novel fluorescent probe CPP30-Lipo/CDKACT4 was developed to monitor CDK4 activity and the therapeutic response of its inhibitor in breast cancer. It demonstrated good optical stability and targetability. The probe successfully monitored CDK4 activity and therapeutic response in living cells and in vivo, providing a new strategy for evaluating antitumor therapeutic efficacy and drug development in a clinical context.
