The development of new materials for biological application requires in vitro testing of cell/surface interactions. Cell adhesion and spreading are difficult to quantify as most materials are non-transparent and transmission microscopy cannot be used. Contrast in reflection microscopy is rather poor. We propose an alternative method for the automated screening of cell attachment and spreading using backscattered electron imaging of scanning electron microscopy. The enhanced cell contrast permits study of cell/material interactions by little differences between cells and material.

Droplet-based vitrification of adherent human induced pluripotent stem cells on alginate microcarrier influenced by adhesion time and matrix elasticity

Summary: The gold standard in cryopreservation is currently the slow freezing method, but there is a need for more sophisticated cryopreservation protocols, especially for hiPSCs, to meet the increasing demand for therapeutic and diagnostic cell types. A novel vitrification-based method for adherent hiPSCs has been developed, with potential applications in suspension-based bioreactors after thawing. By optimizing conditions such as microcarrier stiffness and adhesion time, droplet-based vitrification was found to be superior to conventional slow freezing in preserving cell characteristics and minimizing cell loss.

Compact holographic sound fields enable rapid one- step assembly of matter in 3D

Kai Melde, Heiner Kremer, Minghui Shi, Senne Seneca, Christoph Frey, Ilia Platzman, Christian Degel, Daniel Schmitt, Bernhard Schoelkopf, Peer Fischer

Summary: Acoustic waves can generate forces on matter, allowing for the precise shaping of ultrasound fields in 3D. This technique enables the one-step assembly of particles, hydrogel beads, and biological cells using acoustic forces without the need for opposing waves or support structures. The structures can be fixed by gelation. This advancement holds great potential for tissue engineering and additive manufacturing.
