Summary: This study found that DUSP4 is highly expressed in esophageal squamous cell carcinoma (ESCC) and is negatively correlated with patient prognosis. Knockdown of DUSP4 inhibits cell proliferation and tumor growth, while the loss of Dusp4 in mice significantly inhibits esophageal tumorigenesis. Mechanistically, DUSP4 directly binds to HSP908 and promotes its ATPase activity by dephosphorylating specific sites, affecting the JAK1/2-STAT3 signaling pathway. Treatment with DUSP4 lentivirus or an HSP908 inhibitor impedes PDX tumor growth and inactivates the JAK1/2-STAT3 signaling pathway.

In vitro and in vivo experiments demonstrated that PHGDH knockdown inhibited ESCC progression by suppressing Wnt/beta-catenin signaling pathway. These results indicate that PHGDH may be a potential therapeutic target for ESCC.
