Objective. We hypothesised that the endothelial dysfunction is associated with early glucose dysregulation and/or atherosclerosis risk factors in nondiabetic women with a previous history of gestational diabetes (pGDM). Material/Methods. Anthropometric parameters, glucose regulation (OGTT), insulin resistance (HOMA), lipids, biomarkers of endothelial dysfunction, and inflammation were evaluated in 85 women with pGDM and in 40 controls 2-24 months postpartum. Results. The pGDM group consisted of 67% normoglycemic women (pGDM-N) and 33% with prediabetic state (pGDM-P). The BMI, waist circumference, fasting and 2 h glucose (OGTT), soluble adhesion molecules, tissue plasminogen activator antigen, high sensitivity C-reactive protein, total-, LDL-cholesterol, and triglycerides/HDL-cholesterol ratio were higher in the pGDM women compared with the controls. After adjustment for BMI and fasting glucose, only higher triglycerides, higher TG/HDL and lower HDL-cholesterol were associated with pGDM. The pGDM-P differed from pGDM-N for only higher triglycerides and TG/HDL. The plasma level of sE-selectin was not independently associated with glucose concentration in pGDM group. sE-selectin level correlated with triglycerides, TG/HDL, plasminogen activator inhibitor-1 antigen, and sICAM-1. Conclusions. sE-selectin level correlated with components of metabolic syndrome, but only the atherogenic lipid profile was independently associated with a previous history of GDM in nondiabetic women 2-24 months postpartum.

A Micellar Formulation of Quercetin Prevents Cisplatin Nephrotoxicity

Alfredo G. Casanova, Marta Prieto, Clara I. Colino, Carmen Gutierrez-Millan, Barbara Ruszkowska-Ciastek, Esther de Paz, Angel Martin, Ana I. Morales, Francisco J. Lopez-Hernandez

Summary: This study demonstrated that a micellar formulation of Pluronic F127-encapsulated quercetin (P-quercetin) was more effective in increasing the bioavailability of quercetin and protecting the kidneys in rats when compared to natural quercetin. P-quercetin not only reduced plasma creatinine and urea levels induced by the nephrotoxic drug cisplatin, but also improved some renal function parameters. The new formulation with enhanced properties can potentially lead to the development of quercetin as a nephroprotective agent in clinical settings at lower dosages.

Impact of Adjuvant Treatment on Heparanase Concentration in Invasive, Unilateral Breast Cancer Patients: Results of a Prospective Single-Centre Cohort Study

Barbara Ruszkowska-Ciastek, Kornel Bielawski, Elzbieta Zarychta, Piotr Rhone

Summary: The study found that adjuvant treatment significantly reduced the heparanase levels in invasive breast cancer patients. Higher concentrations of heparanase were noted in estrogen receptor-negative cancers. Additionally, overweight patients and those with larger tumor diameters had a lower chance of lower pre-treatment heparanase concentrations.

Summary: This study examined the utility of SDF-1 alpha and selected vasculo-angiogenic parameters in predicting disease relapse in invasive breast cancer patients. The combination of SDF-1 alpha lower than 42 pg/dL and a number of circulating EPCs lower than 9.68 cells/μL showed strong diagnostic power for predicting disease relapse.

Summary: Using hiPSC technology, researchers derived a subtype of astrocytes from healthy individuals and schizophrenia patients, finding a distinct gene expression signature specific to schizophrenia. Astrocyte diversity may potentially modulate schizophrenia pathophysiology and drug response, warranting further investigation.

DNA Hydroxymethylation in High-Grade Gliomas

Angelika Stapinska-Syniec, Marta Grabiec, Marcin Rylski, Albert Acewicz, Michal Sobstyl

Summary: This review presents the current state of knowledge on DNA hydroxymethylation in human high-grade gliomas. Multiple studies have shown that the expression of 5-hydroxymethylcytosine (5-hmC) and ten-eleven translocation (TET) enzymes is significantly reduced in high-grade gliomas compared to normal brain tissue. Decreased levels of 5-hmC are associated with poor survival, while high expression of the TET3 enzyme is related to a better prognosis for glioblastoma multiforme (GBM) patients. This highlights the importance of DNA hydroxymethylation in molecular diagnostics of human gliomas, including survival estimation and treatment response differentiation.

Cooperation between Angiogenesis, Vasculogenesis, Chemotaxis, and Coagulation in Breast Cancer Metastases Development: Pathophysiological Point of View

Elzbieta Zarychta, Barbara Ruszkowska-Ciastek

Summary: This article summarizes the roles of angiogenesis, chemotaxis, and coagulation in the development of distant metastases in breast cancer. It highlights that breast cancer is still one of the main causes of morbidity and mortality in women worldwide, but the number of deaths has decreased due to improved diagnostic and therapeutic opportunities. However, metastatic disease remains a therapeutic challenge.

The Profiles of Tet-Mediated DNA Hydroxymethylation in Human Gliomas

Summary: Gliomas, the most common malignant brain tumors, are controlled by various epigenetic mechanisms. This study found that the expression of 5-hydroxymethylcytosine (5-hmC) decreases across glioma grades and shows unusual heterogeneity in glioblastoma (GBM). Further analysis revealed that the levels of TET proteins may influence the abundance of 5-hmC in GBM. Additionally, the study suggests that aberrant DNA hydroxymethylation in blood samples may serve as early tumor-related biomarkers for glioma patients.

Summary: The study aimed to investigate the concentration and activity of tissue factor (TF) and tissue factor pathway inhibitor (TFPI) in patients with intracranial neoplasms, as well as the concentration of thrombin-antithrombin (TAT) complexes. The results showed that the concentration and activity of TF were significantly higher in the plasma and tumor tissue-derived homogenate of intracranial tumor patients compared to control individuals. In contrast, the concentration of TFPI was lower in the tumor tissue-derived homogenate. These findings suggest an increased prothrombotic risk in intracranial tumor patients due to the imbalance between TF and TFPI.

Summary: Vitamin D deficiency is a significant issue in the global population, especially among older people. Adipose tissue is the main storage site for vitamin D and its regulation affects adipogenic genes and adipocyte apoptosis, as well as the secretion of appetite-regulating hormone-leptin. Leptin, visfatin, and ghrelin are found to be associated with vitamin D concentration and eGFR rate in geriatric females.

Summary: The study evaluated the number of endothelial progenitor cells (EPCs) in the blood of patients with Psoriasis vulgaris (PsV) and found that PsV patients had significantly higher levels of EPCs compared to healthy individuals, with the levels inversely correlated with disease severity. No significant associations were found between EPC count and other factors such as age, body mass index, gender, and smoking habits.
