Magnetic chelating nanoprobes for enrichment and selective recovery of metalloproteases from human saliva

New magnetic nanoprobes based on surface-functionalized magnetite particles were synthesized and used to selectively capture metalloproteases from human saliva samples. In addition to their high specific surface area, these nanoparticles have metal ion chelating moieties grafted on their surface by the reaction with the organosilane EDTA-TMS ((N-(trimethoxysilylpropyl)-ethylenediaminetriacetate trisodium salt). The most distinct feature of these particles is their capability to selectively recover metalloproteases even in highly diluted saliva samples. The high affinity of these materials for metalloproteases was attributed to the formation of chelates between the chelating moieties and the metal ions of metalloproteases. These magnetic particles exhibited high colloidal stability in the biological standard buffers MES, HEPES, TEAB and Tris-HCl, within a significant pH range (pH 5-9) and, due to their magnetic features, enabled a fast separation of the metalloproteases from the medium by simply applying an external magnetic field. Thus, these materials were proven to be valuable probes for the selective enrichment and rapid recovery of metalloproteases from human saliva, particularly when dealing with trace amounts of material.

This article provides an overview of the mechanistic uptake/release of these compounds, discusses the potential ecological consequences, and explores analytical methodologies for their determination. It also highlights the limitations of current literature and presents future research prospects.

Transition metal-modification of carrageenan-silica hybrids by a sol-gel method

Summary: This study demonstrates the successful modification of hybrid siliceous materials with transition metals (TM = Co2+, Ni2+, Cu2+, Zn2+) using a sol-gel method with the polysaccharide kappa-carrageenan as a precursor. Structural analysis and characterization studies were conducted, and the effects of the TM ions on the properties of the hybrids were assessed. The results show that the modification with TM induces changes in the morphological, optical, and thermal properties of the hybrids compared to the unmodified analogs, indicating the practical applicability of the sol-gel method for modifying the properties of host particles.

Microplastics in Sediments from a Sandy Beach in Costa Nova (Aveiro, Portugal)

Summary: The main objective of this study was to estimate the microplastic pollution on Costa Nova sandy beach in Portugal using longitudinal and cross-sectional transects. The results showed low variability in data, with median concentrations ranging from 142 to 356 p/kg d.w. in cross-sectional transects, and from 211 to 270 p/kg d.w. in longitudinal transects. Microspheres were the predominant morphology, accounting for over 90% of all samples, and the most abundant microplastics were <200 μm in size. Despite moderate-low tourist activity, microplastic contamination was higher in Costa Nova compared to other Portuguese beaches, possibly due to nearby port activities and ocean currents. However, this study highlights the disparity in data caused by different methodologies used in analyzing similar areas.

Several differentially expressed proteins related to the diseases were identified. Machine learning analysis confirmed the potential of the salivary proteome to discriminate AIH and PBC patients. The functional enrichment analysis highlighted the involvement of the immune system, liver fibrosis process, and antimicrobial activity.

Expression of NORAD correlates with breast cancer aggressiveness and protects breast cancer cells from chemotherapy

Summary: The recently discovered lncRNA NORAD is induced after DNA damage in a p53-dependent manner and it plays a critical role in maintaining genomic stability. NORAD is upregulated in several types of cancer, including breast cancer, and high NORAD expression levels are associated with poor prognosis. In this study, it was found that downregulation of NORAD sensitizes triple-negative breast cancer cells to chemotherapy.

A two-pronged approach against glioblastoma: drug repurposing and nanoformulation design for in situ-controlled release

Summary: This study proposes a two-pronged approach for tackling glioblastoma (GB) using drug repurposing with celecoxib (CXB) and a nanoformulation with ultra-small nanostructured lipid carriers (usNLCs). The results demonstrate that by enhancing the design and construction of the nanoformulation, the solubility and loading properties of CXB can be improved, enabling controlled drug release and high cytotoxicity. This nanoformulation holds promise for improving the treatment of GB.

Gold nanoparticle probes for colorimetric detection of plasma galectin-3: a simple and rapid approach

Summary: In this study, a colorimetric and low-cost approach for detecting and quantifying Gal-3 using gold nanoparticles bioconjugated with Gal-3 antibody was developed. The method showed a linear response of the absorbance ratio to Gal-3 concentration, accompanied by a change in color intensity. It provided faster results and was more cost-effective compared to conventional detection techniques.
