AIM: To study the levels of serum soluble intercellular adhesion molecule-1 (sICAM-1), plasma D-lactate and diamine oxidase (DAO) in patients with inflammatory bowel disease (IBD), and the potential clinical significance. METHODS: Sixty-nine patients with IBD and 30 healthy controls were included in this study. The concentration of sICAM-1 was detected with enzyme-linked immunosorbent assay, the level of D-lactate and DAO was measured by spectroscopic analysis, and the number of white blood cells (WBC) was determined by routine procedure. RESULTS: The levels of sICAM-I, DAO, and WBC in IBD patients were significantly higher than those in the control group (P < 0.01). sICAM-1 in IBD patients was found to be closely related to the levels of DAO and D-lactate (212.94 +/- 69.89 vs 6.35 +/- 2.35, P = 0.000), DAO (212.94 +/- 69.89 vs 8.65 +/- 3.54, P = 0.000) and WBC (212.94 +/- 69.89 vs 7.40 +/- 2.61, P = 0.000), but no significant difference was observed between patients with ulcerative colitis and patients with Crohn's disease. The post-treatment levels of sICAM-1, D-lactate and WBC were significantly lower than before treatment (sICAM-I 206.57 +/- 79.21 vs 146.21 +/- 64.43, P = 0.000), (D-lactate 1.46 +/- 0.94 vs 0.52 +/- 0.32, P = 0.000) and (WBC 7.24 +/- 0.2.33 vs 5.21 +/- 3.21, P = 0.000). CONCLUSION: sICAM-1, D-lactate and DAO are closely related to the specific conditions of IBD, and thus could be used as a major diagnostic index. (c) 2009 The WJG Press and Baishideng. Summary: This study aimed to establish a scoring model based on early brain injury indicators to predict the functional outcomes of patients with aneurysmal subarachnoid hemorrhage (aSAH) at 90 days. The model, named TAPS, showed good discrimination and calibration in both the derivation and validation cohorts, and proved to be an easy handle tool for clinicians to predict the 90-day outcomes and identify patients at risk for poor prognosis.

The Added Value of Second-Look Ultrasound in Patients With Primary Hyperparathyroidism With Discordant or Negative Previous Imaging Findings

Siqi Jin, Qingli Zhu, Li Huo, Ou Wang, Xiaoping Xing, Quan Liao, Li Ma, Jianchu Li, Yuxin Jiang, He Liu

Summary: The study aimed to evaluate the value of second-look ultrasound (US) for preoperative localization in patients with primary hyperparathyroidism with negative or discordant imaging results. The sensitivity, positive predictive value, and accuracy of second-look US were found to be 92.1%, 95.1%, and 88.1%, respectively. Overall, second-look US is a promising method for preoperative localization, especially for patients with negative or discordant imaging findings.

Application of ddPCR in detection of the status and abundance of EGFR T790M mutation in the plasma samples of non-small cell lung cancer patients

Hui Zhang, Yi Hu, Yan Wang, Xia Song, Ying Hu, Li Ma, Xinjie Yang, Kun Li, Na Qin, Jinghui Wang, Jialin Lv, Xi Li, Xinyong Zhang, Quan Zhang, Yuhua Wu, Guangyin Yao, Shucai Zhang

Summary: This study assessed the performances of droplet digital polymerase chain reaction (ddPCR) and next-generation sequencing (NGS) in detecting EGFR T790M mutation in the plasma samples of NSCLC patients. The results showed that ddPCR had a high sensitivity and specificity in detecting EGFR T790M mutation. Furthermore, a higher EGFR T790M abundance was associated with a better response to third-generation EGFR-TKIs.

New Methodology for Determining Plasma Protein Binding Kinetics Using an Enzyme Reporter Assay Coupling with High-Resolution Mass Spectrometry

Zhengyin Yan, Li Ma, Julie Huang, Pasquale Carione, Jane R. Kenny, Cornelis E. C. A. Hop, Matthew Wright

Summary: Determination of drug binding kinetics in plasma is challenging, and this study introduces a new parameter called dynamic free fraction to describe drug-protein binding kinetics. The dynamic free fraction can be determined by coupling the drug binding assay with a reporter enzyme and high-resolution mass spectrometry. This methodology bypasses the challenges in existing methods and allows assessment of the impact of protein binding kinetics on drug properties.

Bispecific anti-CD3xanti-CD155 antibody mediates T-cell immunotherapy in human haematologic malignancies

Li Ma, Juan Ma, Xin Sun, Honggang Liu

Summary: T cells play a crucial role in the cell-mediated antitumour response. Bispecific antibodies (Bi-Abs) have shown promise in recruiting T cells for killing tumours. This study demonstrates the expression of CD155 in a wide range of human haematologic tumours and highlights the effectiveness of the bispecific antibody anti-CD3 x anti-CD155 in activating T cells to target malignant haematologic cells. The results show that CD155Bi-Ab-armed T cells exhibit enhanced cytotoxicity and increased secretion of cell-killing mediators and T-cell-derived cytokines, indicating that CD155 could be a novel target for immunotherapy against haematologic malignancies.

Association of embolization with long-term outcomes in brain arteriovenous malformations: a propensity score-matched analysis using nationwide multicenter prospective registry data

Summary: This study compared the long-term outcome of hemorrhagic stroke or death in patients with conservative management or stand-alone embolization for AVM. The results showed that embolization did not prevent long-term hemorrhagic stroke or death compared with conservative management in the overall cohort, as well as in the unruptured and ruptured AVM subgroups. Stratified analysis suggested that target embolization might be beneficial for unruptured AVMs, while curative embolization improved the outcomes of ruptured AVMs. The long-term neurological status was similar between the two strategies.

Optic Fiber Microsensor Reveals Specific Spatiotemporal Oxygen Uptake Profiles at the Mammalian Ocular Surface

Qin Sun, Li Ma, Fernando Ferreira, Chelsea Brown, Brian Reid, Min Zhao

Summary: The uptake of oxygen by cells and tissues is an important indicator of metabolic demand, microenvironment changes, and pathophysiology. Using a non-invasive optical fiber oxygen sensor, researchers have revealed a spatiotemporal profile of oxygen uptake at the ocular surface of rodents and non-human primates. The results showed a distinct oxygen uptake gradient in mice, with a higher influx at the limbus and conjunctiva regions compared to the center of the cornea. The findings provide valuable insights for comparative studies and understanding the response of the cornea and other tissues to various insults or environmental changes.

PAI-1 regulates AT2-mediated re-alveolarization and ion permeability

Gibran Ali, Mo Zhang, Jianjun Chang, Runzhen Zhao, Yang Jin, Jiwang Zhang, Hong-Long Ji

Summary: This study finds that elevated levels of PAI-1 are present in both lung edema fluid and the circulating system in patients with acute lung injury. The study also reveals that PAI-1 inhibits the self-renewal of alveolar epithelial cells (AT2), thereby affecting the regeneration of alveolar epithelium. Through the investigation of a humanized mouse line carrying Serpine1 gene mutants, it is found that Serpine1 gene mutations lead to a decrease in the number of AT2 cells and a delay in the formation of injured epithelium.

Summary: In this study, quantitative digital subtraction angiography (QDSA) was used to explore the hemorrhagic quantitative hemodynamic indicators of brain arteriovenous malformation (AVM) nidus. The results showed that a higher stasis index and slower transnidal relative velocity (TRV) were significantly correlated with AVM rupture and could be used as effective indicators to predict future hemorrhage risk.
