Increased expression of fractalkine and its receptor CX3CR1 in canine inflammatory bowel disease and their possible role in recruitment of intraepithelial lymphocytes

The interaction between fractalkine/CX(3)CL1 and its receptor CX(3)CR1 has been reported to play an important role in various human inflammatory diseases, including inflammatory bowel disease (IBD) mediated by lymphocyte chemoattraction. The objective of this study was to investigate the role of fractalkine and CX(3)CR1 in lymphocyte migration in canine IBD. IBD was diagnosed in 34 dogs, and 19 healthy beagles were used as normal controls. We quantified intestinal mRNA and protein expression of fractalkine and CX(3)CR1 by real-time RT-PCR and ELISA, respectively, and examined the localization of fractalkine in canine intestine by immunohistochemistry. The expression of CX(3)CR1 and surface antigens on peripheral blood mononuclear cells (PBMCs) and intraepithelial lymphocytes (IELs) was analyzed by flow cytometry. Intestinal fractalkine and CX(3)CR1 mRNA was significantly up-regulated in IBD dogs compared with the healthy control dogs. In addition, fractalkine expression on intestinal epithelial cells was significantly increased in the intestinal mucosa of IBD dogs compared with the healthy dogs. CX(3)CR1(+) PBMCs were significantly elevated in IBD dogs and positively correlated with the histopathological severity of IELs infiltration. These CX(3)CR1(+) PBMCs predominantly expressed markers for cytotoxic T cells. Almost all IELs expressed CD3, and the majority of cells expressed CD8 rather than CD4, which was analogous to the CX(3)CR1(+) PBMCs. These results suggest that the fractalkine-CX(3)CR1 interaction may contribute to the pathogenesis of canine IBD through migration of IELs. (c) 2012 Elsevier B.V. All rights reserved.

Summary: Regulatory T cells recruitment in canine urothelial carcinoma is associated with the BRAF(V595E) mutation and tumor-produced CCL17. Elevated CCL17 mRNA expression in bladder and increased CCL17 protein concentration in urine were found in dogs with urothelial carcinoma. The presence of higher levels of tumor-infiltrating regulatory T cells and urine CCL17 concentration were correlated with a poor prognosis in these dogs.

Efficacy and adverse events of continuous l-asparaginase administration for canine large cell lymphoma of presumed gastrointestinal origin

Anaphylaxis after vaccination for cats in Japan

Association between intestinal lymphangiectasia and expression of inducible nitric oxide synthase in dogs with lymphoplasmacytic enteritis

Summary: Intestinal lymphangiectasia (IL) is a common complication in dogs. This study aimed to investigate the potential role of inducible nitric oxide synthase (iNOS) in the development of IL in dogs with lymphoplasmacytic enteritis (LPE). The results showed that there was a significant increase in iNOS expression in the duodenal tissues of dogs with IL-negative or IL-positive LPE compared to control dogs. However, there was no significant difference in iNOS expression between IL-positive and IL-negative tissues, suggesting that factors other than nitric oxide may contribute to the development of IL in dogs with LPE.

Retrospective study of feline tracheal mass lesions

Summary: This multicentre, retrospective observational study aimed to describe the clinical presentation, diagnostic methods, treatment and outcomes of cats with tracheal masses. Lymphoma was the most prevalent diagnosis, and showed a good response to chemotherapy with or without radiation therapy. Ultrasound-guided fine-needle biopsy and cytology are good diagnostic procedures for cervical tracheal lesions.

Serum level of apoptosis inhibitor of macrophage in dogs with histiocytic sarcoma and its association with the disease

Summary: Histiocytic sarcoma (HS) is a rare malignancy in dogs, with poor prognosis. This study investigated the involvement of apoptosis inhibitor of macrophage (AIM) in the development of HS in dogs. The expression of AIM in tumor tissue and serum AIM levels were evaluated, and the effects of AIM overexpression on HS cells were studied. The results suggest that elevated expression of AIM in tumor and serum could induce the progression of HS cells in dogs, and serum AIM levels may serve as a biomarker for HS.
