Summary: This study found that macrophages are the main source of IL-31 in atopic dermatitis (AD) skin lesions. It demonstrated the association of macrophages, TSLP, and periostin with AD itch, and the attenuation of itch symptoms with macrophage depletion. The study suggests that a network comprising IL-31-expressing macrophages, TSLP, periostin, and basophils plays a significant role in AD itch.

Summary: TSLP, particularly the long form, plays a crucial role in immune-mediated skin diseases, driving Th2 and Th17 immune responses, inducing itch, and being associated with skin fibrosis and epidermal hyperplasia. Targeting TSLP may have significant implications for the treatment of these diseases in the future.

Summary: This study found that selective HIF prolyl hydroxylase inhibitors can suppress TSLP production in allergic skin diseases, suggesting their potential as antiallergic drugs. Controlling the HIF activation system may have therapeutic potential in atopic dermatitis.

Summary: Our study found that vitamin D3 can induce skin dendritic cells to differentiate into Th2 cells producing IL-4 and IL-13, while also triggering the release of TSLP. Blocking the TSLP receptor in some donors eliminated IL-13 production but had no effect on IL-4+ Th2 cells. Vitamin D3-elicited CD14+ dendritic cells were able to promote IL-4+ Th2 cells in a TSLP-independent manner.

Summary: Canine atopic dermatitis (cAD) is a chronic and recurrent inflammatory skin disorder. Allergen-specific immunotherapy (ASIT) is recognized as an effective treatment for allergies. This study analyzed changes in gene expression in cAD patients after undergoing ASIT. Nine differentially expressed genes related to the dysregulated immune response in cAD patients were identified, and the expressions of five of these genes changed after ASIT.

Summary: TSLP is a critical cytokine in allergy and asthma development, primarily produced by epithelial cells. It functions as a potent immune system activator through interaction with its receptor complex. Understanding the interplay between TSLP and dysfunctional epithelium provides insights into allergy and asthma pathogenesis.

Summary: This study demonstrates the potential of immunotherapy for lung cancer by examining the impact of Thymic Stromal Lymphopoietin (TSLP) cytokine induction on early lung cancer development. TSLP induction suppresses the development of invasive lung tumors, dependent on CD4(+) T cells. This highlights the importance of adaptive immune response in protecting against lung cancer progression.

Summary: This study found that treatment with poly (I:C) and exogenous activation of TLR3 can exacerbate calcipotriol-induced atopic dermatitis (AD)-like skin lesions in mice, possibly by increasing the production of TSLP and other Th2 cytokines.

Summary: Allergic sensitisation in the skin can lead to allergic dermatitis and further to airway asthma. This study examines the difference between superficial or deep skin sensitisation, characterises the immune cells generated, and shows differential TSLP and IL-1 beta involvement.

Summary: Chloroquine (CQ) exhibits anti-inflammatory effects by modulating TSLP levels in mast cells and reducing ear edema thickness, as well as TSLP levels in ear tissue and serum. The down-regulation of CASP1, NF-kappa B, and other signaling cascades by CQ contributes to its protective role against TSLP-mediated inflammatory disorders.

Summary: Two isoforms of TSLP, lTSLP and sTSLP, exist in rheumatoid arthritis (RA), with lTSLP being predominant in active RA and sTSLP in inactive RA and healthy controls. lTSLP expression is associated with disease activity and proinflammatory cytokine expression. lTSLP promotes the expression of proinflammatory cytokines in PBMCs of RA, while inhibiting the expression of anti-inflammatory cytokines.

Summary: As indicated by the study, similar pandemics may occur more frequently in the future due to factors such as climate change, consumption of exotic foods and drinks, global travel, increasing world population, decreasing production space, declining wildlife and free-ranging animal habitats, as well as changed lifestyles. Both society and health authorities need to be alert and well-prepared for similar future situations, with research focusing on prevention and rapid development of treatment options (medications, vaccines).
