Sensitive Detection of Thirteen Bacterial Vaginosis-Associated Agents Using Multiplex Polymerase Chain Reaction

Bacterial vaginosis (BV) is characterized by a polymicrobial proliferation of anaerobic bacteria and depletion of lactobacilli, which are components of natural vaginal microbiota. Currently, there are limited conventional methods for BV diagnosis, and these methods are time-consuming, expensive, and rarely allow for the detection of more than one agent simultaneously. Therefore, we conceived and validated a multiplex polymerase chain reaction (M-PCR) assay for the simultaneous screening of thirteen bacterial vaginosis-associated agents (BV-AAs) related to symptomatic BV: Gardnerella vaginalis, Mobiluncus curtisii, Mobiluncus mulieris, Bacteroides fragilis, Mycoplasma hominis, Atopobium vaginae, Ureaplasma urealyticum, Megasphaera type I, Clostridia-like bacteria vaginosis-associated bacteria (BVABs) 1, 2, and 3, Sneathia sanguinegens, and Mycoplasma genitalium. The overall validation parameters of M-PCR compared to single PCR(sPCR) were extremely high, including agreement of 99.1% and sensitivity, specificity, and positive predictive values of 100.0%, negative predictive value of 97.0%, accuracy of 99.3%, and agreement with Nugent results of 100.0%. The prevalence of BV-AAs was very high (72.6%), and simultaneous agents were detected in 53.0%, which demonstrates the effectiveness of the M-PCR assay. Therefore, the M-PCR assay has great potential to impact BV diagnostic methods in vaginal samples and diminish associated complications in the near future.

Summary: This study analyzed the immune response of patients with SARS-CoV-2 infection and identified biomarkers related to disease severity. Some angiogenesis markers were more frequently elevated in severe cases compared to other groups. In terms of cardiovascular diseases, D-dimer, GDF-15, and sICAM-1 were higher in the severe cases. Significant differences in cytokines and chemokines levels were also observed according to the clinical course.

Summary: Cervical cancer is a major health issue in Latin America and Caribbean, and many countries have not been successful in implementing screening programs. Self-sampling, as an alternative approach, has shown promise. This systematic review summarizes recent HPV self-sampling approaches in the region and highlights the potential of this strategy in overcoming barriers to screening and increasing attendance among underscreened women.

The combination improved the overall effectiveness of conventional drugs, increasing cytotoxicity, antimigratory effects, and reversing resistance. Combining conventional drugs with bioactive compounds derived from cinnamic acid could provide new treatment options for different types of breast cancer.

Summary: Cervical cancer is a leading cause of death among women, and current treatment options for endocervical adenocarcinoma (ECA) are ineffective. This study evaluated the effectiveness of hypericin (HYP) encapsulated on Pluronic F127 (F127/HYP) photodynamic therapy (PDT) on human cell lines derived from invasive cervical adenocarcinoma (HeLa) compared to a human epithelial cell line (HaCaT). The results showed that F127/HYP had a dose-dependent and selective phototoxic effect on HeLa cells, caused cell death through necrosis and reactive oxygen species (ROS) production, and reduced long-term proliferation and migration capacity of HeLa cells. These findings suggest that F127/HYP micelles may be a promising approach for specifically treating ECA using PDT.
