Evaluation of Hemagglutination Activity of Chitosan Nanoparticles Using Human Erythrocytes

Chitosan is a polysaccharide composed of randomly distributed chains of beta-(1-4) D-glucosamine and N-acetyl-D-glucosamine. This compound is obtained by partial or total deacetylation of chitin in acidic solution. The chitosan-based hemostatic agents have been gaining much attention in the management of bleeding. The aim of this study was to evaluate in vitro hemagglutination activity of chitosan nanoparticles using human erythrocytes. The preparation of nanoparticles was achieved by ionotropic gelification technique followed by neutralization with NaOH 1mol/L-1. The hemagglutination activity was performed on a solution of 2% erythrocytes (pH 7.4 on PBS) collected from five healthy volunteers. The hemolysis determination was made by spectrophotometric analysis. Chitosan nanoparticle solutions without NaOH addition changed the reddish colour of the wells into brown, suggesting an oxidative reaction of hemoglobin and possible cell lysis. All neutralized solutions of chitosan nanoparticles presented positive haemagglutination, without any change in reaction color. Chitosan nanoparticles presented hemolytic activity ranging from 186.20 to 223.12%, while neutralized solutions ranged from 2.56 to 72.54%, comparing to distilled water. Results highlight the need for development of new routes of synthesis of chitosan nanoparticles within human physiologic pH.

Summary: The study presents the synthesis of a novel series of pyridine-thiazolidinones with anti-Trypanosoma cruzi and leishmanicidal activities. In vitro assays showed that several compounds demonstrated inhibitory effects on trypomastigotes and amastigotes of Trypanosoma cruzi, as well as on promastigotes and amastigotes of Leishmania amazonensis. Compound 27 displayed higher selectivity against amastigote form compared to Miltefosine. Overall, the results suggest that these pyridine-thiazolidinones could serve as potential starting points for the development of new antiparasitic agents.

Summary: This study focused on increasing the aqueous solubility of LPSF/GQ-238 by obtaining solid dispersions using different carriers. The results showed that the higher the proportion of polymer used in the dispersion, the greater the solubility observed. Morphological and spectroscopic analyses demonstrated the conversion of the crystalline structure of the molecule into an amorphous state in the dispersions. The in vitro study revealed improved schistosomicidal activity of the dispersed NCE against adult worms and schistosomulae.

In vitro evaluation of alkaline lignins as antiparasitic agents and their use as an excipient in the release of benznidazole

Summary: The Amazon rainforest is the world's largest tropical timber reserve, and the management of native forests in the region is a highly sensitive geopolitical issue. This study extracted and characterized lignins from the branches and leaves of Protium puncticulatum and Scleronema micranthum, and evaluated their antioxidant potential, cytotoxicity against animal cells, and antiparasitic activity against Leishmania amazonensis, T. cruzi, and chloroquine-sensitive and resistant Plasmodium falciparum. The lignins showed potential as antioxidants and had low cytotoxicity, and they were able to inhibit the growth of promastigotes and trypomastigotes in vitro. However, their activity against Plasmodium falciparum was limited, and they did not inhibit the growth of Schistosoma mansoni parasites. Overall, the findings highlight the potential of these understudied lignins for various biological activities.

Oral Mucositis in Children with Leukemia Undergoing Chemotherapy: A Case Series

The severity of oral mucositis and hematological parameters fluctuated during the follow-up, and all patients were exposed to stomatotoxic drugs during the initial phase of cancer treatment.

Summary: In this study, poly(lactic acid)/poly(ethylene glycol) fibers were produced for the controlled release of acyclovir, a drug used in the treatment of oral recurrent herpes. The results showed that the addition of hydrophilic poly(ethylene glycol) improved the drug release, and the fibers were found to be non-toxic to cells.
