Overexpression of response gene to complement 32 (RGC32) promotes cell invasion and induces epithelial-mesenchymal transition in lung cancer cells via the NF-κB signaling pathway

Response gene to complement 32 (RGC32) is a novel cellular protein that has been reported to be expressed aberrantly in multiple types of human tumors. However, the role of RGC32 in cancer is still controversial, and the molecular mechanisms by which RGC32 contributes to the development of cancer remain largely unknown. In the present study, we constructed a recombinant expression vector pCDNA3.1-RGC32 and transfected it into human lung cancer A549 cells. Stable transformanted cells were identified by real-time PCR and Western blot analysis. Functional analysis showed that forced overexpression of RGC32 increased invasive and migration capacities of lung cancer cells in vitro, and induced the acquisition of epithelial-mesenchymal transition (EMT) phenotype, as demonstrated by the spindle-like morphology, downregulation of E-cadherin, and upregulation of Vimentin, Fibronectin, Snail and Slug. Also, overexpression of RGC32 increased expression and activities of matrix metalloproteinase (MMP)-2 and MMP-9 in A549 cells. Furthermore, the downregulation of E-cadherin induced by RGC32 was remarkably attenuated by nuclear factor-kappa B (NF-kappa B) inhibitor BAY 11-7028 and small interfering RNA targeting NF-kappa B p65, suggesting a role of the NF-kappa B signaling pathway in RGC32-induced EMT. Taken together, our data suggest that RGC32 promotes cell migration and invasion and induces EMT in lung cancer cells via the NF-kappa B signaling pathway.

Summary: This study collected nasopharyngeal swabs, anal swabs, saliva, blood, and urine specimens from patients with a course of disease ranging from 7 to 69 days. It was found that the positive detection rate was highest in nasopharyngeal swabs, followed by anal swabs. There was no significant correlation between antibody levels and days after symptom onset or viral load.

Summary: Strongly immunogenic neoantigens were identified from NSCLC patients through whole-exome sequencing and machine-learning-based predictions. The efficient antitumor responses of NRTs were demonstrated in transgenic mice and tumor-bearing mouse models both in vivo and in vitro. These results suggest the feasibility of developing neoantigen-based personalized immunotherapies for NSCLC.

Summary: This study successfully constructed a diagnostic model for lung cancer using the 5hmC signal and fragmentation profiles in 5hmC cfDNA data, which can effectively distinguish lung cancer patients from healthy individuals. The models showed good performance in different lung cancer subtypes and stages, and were associated with patient survival rates. The results demonstrate the significant application value of clinical diagnostic models based on cfDNA 5hmC data.

Adoptive neoantigen-reactive T cell therapy: improvement strategies and current clinical researches

Summary: Neoantigens generated by non-synonymous mutations can activate neoantigen-reactive T cells and effectively inhibit tumor growth. Immunotherapy based on these T cells has shown great success in treating melanoma and other solid tumors. Improvements in the preparation and function evaluation of these T cells, as well as ongoing clinical trials, are discussed in this review.
