In conifers, vegetative propagation of superior genotypes is the most direct means for making large genetic gains, because it allows a large proportion of genetic diversity to be captured in a single cycle of selection. There are two aims of vegetative propagation, namely large-scale multiplication of select genotypes and production of large numbers of plants from scarce and costly seed that originates from controlled seed orchard pollinations. This can be achieved, in some species, either through rooted cuttings or rooted microshoots, the latter regenerated through tissue culture in vitro. Thus far, both strategies have been used but often achieved limited success mainly because of difficult and inefficient rooting process. In this overview of technology, we focus on the progress in defining the physical and chemical factors that help the conifer cuttings and microshoots to develop adventitious roots. These factors include plant growth regulators, carbohydrates, light quality, temperature and rooting substrates/media as major variables for development of reliable adventitious rooting protocols for different conifer species.

Monitoring system and in situ conservation of endemic and threatened Beta patula Aiton populations in Madeira Region

H. Nobrega, G. Freitas, M. A. Zavattieri, C. Ragonezi, L. Frese, M. A. A. Pinheiro de Carvalho

Summary: The research conducted on Beta patula species in Madeira Archipelago included a complete floristic survey on two islands to understand the population dynamics and ecological requirements, as well as calculating biodiversity indices to assess the vegetation. The demographic status of Beta patula populations on the islands was determined yearly between 2014 and 2018, providing data for the establishment of a protocol to monitor and manage a genetic reserve for Beta patula and other crop wild relatives. This work aims to contribute to the implementation of the European genetic reserve network.

Review of Sewage Sludge as a Soil Amendment in Relation to Current International Guidelines: A Heavy Metal Perspective

Nuno Nunes, Carla Ragonezi, Carla S. S. Gouveia, Miguel A. A. Pinheiro de Carvalho

Summary: Sewage sludge, an underused biomass derived from wastewater treatment plants, can be utilized as biofertilizer after proper stabilization due to its high content of inorganic matter, nitrate, and phosphorus. However, a wide range of pollutants may be present in sewage sludge, which could have detrimental effects on the environment with long-term applications. Proper screening for heavy metals is essential when utilizing sewage sludge, along with adherence to guidelines and regulations for its application.

Summary: This study summarizes the in situ and ex situ conservation research on the critically endangered species Beta patula, focusing on the associated plant communities. Valuable data was obtained through vegetation census on uninhabited islets Desembarcadouro and Chao, which will contribute to the implementation of conservation actions and management protocols. This work is part of the EU LIFE Project LIFE RECOVER NATURA and was conducted by members of the ISOPlexis Center at the University of Madeira.

Viral diagnosis in cultivars of Ipomoea batatas (L.) Lam.

Carlos G. Andrade, Emanuel M. da Silva, Carla Ragonezi, Miguel A. A. Pinheiro De Carvalho

Summary: The study aimed to screen Begomovirus, Potyvirus, and Carlavirus infections in 16 traditional sweet potato cultivars. The infection prevalence among cultivars was high, with some showing vulnerability to multiple viral infections. A new divergent strain of the SPCFV species was identified in one of the accessions.

Eucalyptus Field Growth and Colonization of Clones Pre-Inoculated with Ectomycorrhizal Fungi

Lidiomar Soares da Costa, Paulo Henrique Grazziotti, Arley Jose Fonseca, Debora Cintia dos Santos Avelar, Marcio Jose Rossi, Enilson de Barros Silva, Eliane Cristine Soares da Costa, Danielle Cristina Fonseca Santos Grazziotti, Carla Ragonezi

Summary: Ectomycorrhizae are classified as a biotechnology to increase the sustainability of planted forests, with fieldwork needed to confirm effectiveness. Inoculation of Eucalyptus clones with ectomycorrhizal fungi in the nursery boosts early plant growth after planting in the field, depending on the specific fungus and clone. Ectomycorrhizal colonization of Eucalyptus occurs naturally and increases with tree development in the field.

Miguel A. A. Pinheiro de Carvalho, Carla Ragonezi, Maria Cristina O. Oliveira, Fabio Reis, Fabricio Lopes Macedo, Jose G. R. de Freitas, Humberto Nobrega, Jose Filipe T. Gananca

Summary: The Climate-Madeira Strategy (CMS) predicts climate scenarios for the Madeira region in 2050 and 2070, including an increase in average temperature and a decrease in precipitation. This study establishes a baseline for a typical agricultural system in Madeira and investigates the impacts of climate change on its components and productivity.

Distribution of Genetic Diversity in Beta patula Aiton Populations from Madeira Archipelago, Portugal

Carla Ragonezi, Humberto Nobrega, Maria Ines Leite, Jose G. R. de Freitas, Fabricio Lopes Macedo, Miguel A. A. Pinheiro de Carvalho

Summary: This study presents the spatial distribution of genetic variability and diversity in B. patula populations using eight polymorphic SSR markers. Despite some genetic differences, the CI and DI populations have more similarities than differences. Analysis of the Molecular Variance showed that 9% of the diversity is between populations, 68% among individuals, and 23% within individuals.

Estimation of Productivity and Above-Ground Biomass for Corn (Zea mays) via Vegetation Indices in Madeira Island

Fabricio Lopes Macedo, Humberto Nobrega, Jose G. R. de Freitas, Carla Ragonezi, Lino Pinto, Joana Rosa, Miguel A. A. Pinheiro de Carvalho

Summary: The advancement of technology in the field, especially the use of UAVs and multispectral cameras, allows real-time monitoring of crop conditions and contributes to machine learning. This study aimed to estimate corn crop productivity and above-ground biomass using different vegetation indices from high-resolution aerial imagery. Strong correlations were found between productivity and the NDVI (p < 0.05; 0.719), as well as the NDRE and GNDVI with crop productivity (p < 0.01; 0.809 and 0.859). The results for above-ground biomass were consistent with previous findings, with high correlations for GNDVI and NDRE (p < 0.05; 0.758 and 0.695). The GNDVI and NDRE indices showed good coefficients of determination for productivity and above-ground biomass estimation. Predicting crop productivity is crucial for producers, allowing them to make timely decisions and contribute to AI integration for drone optimization.

Assessing the Diversity of Sea Beet (Beta vulgaris L. ssp. maritima) Populations

F. Ascarini, H. G. M. Nobrega, I. S. Leite, G. Freitas, C. Ragonezi, M. Amely Zavattieri, M. A. A. Pinheiro de Carvalho

Summary: Sea beet, a morphologically and genetically variable species, is a valuable genetic resource for resistance improvement in beets. Characterization of Madeiran sea beet populations revealed high quantitative variation among populations, with plant height and inflorescence height parameters having the greatest influence on population separation. Molecular analysis using SSRs showed genetic variability between populations, with PCA and PCoA identifying three clusters based on morphological and genetic traits, highlighting the influence of specific environmental conditions on genetic variability and the potential for these populations to be gene sources for sugar beet breeding programs.
