Summary: The immune response to an allograft can activate lymphocytes that cause rejection. The activation of T regulatory cells can reduce allograft rejection and induce immune tolerance. Activated T regulatory cells can be distinguished by various markers. A more detailed characterization of these cells may help reduce non-specific immunosuppression.

Summary: The intestinal immune system must tolerate food antigens to avoid allergy, a process requiring CD4(+) T cells. Combining antigenically defined diets with gnotobiotic models, the study shows that both food and microbiota have distinct effects on the profile and T cell receptor repertoire of intestinal CD4(+) T cells. The response of CD4(+) T cells to food is disrupted by inflammatory challenge, but protection against food allergy is associated with Treg clonal expansion and decreased proinflammatory gene expression.

Summary: This study revealed quantitative and functional B cell defects in multiple myeloma patients undergoing auto-HSCT, leading to compromised immune defense. Stimulation with CD40L/IL-21 partially restored TD B cell function, indicating potential extrinsic drivers of the functional B cell defect. Testing CD4+, CD19+, memory B cells, and MZ-like B cells as biomarkers in prospective studies is suggested.

Summary: This study compared the reconstitution of NK cells and T cells in alloSCT recipients receiving preemptive therapy or letermovir prophylaxis, in order to identify potential biomarkers predicting prolonged and symptomatic HCMV reactivation. Results showed that letermovir prophylaxis prevented HCMV reactivation and increased the numbers of HCMV-specific CD4+ and CD8+ T cells. Low frequencies of HCMV-specific CD4+ T cells and high frequencies of regulatory T cells (Treg) were identified as significant predictors of prolonged and symptomatic HCMV reactivation.

Summary: The accumulation of DNA mutations in aging hematopoietic stem cells (HSCs) increases the expression of MHCII on their surface, allowing them to be recognized by bone marrow regulatory T cells (Tregs). Tregs transfer cAMP to aged HSCs through gap junctions, promoting their survival and preventing apoptosis. Disrupting the interaction between HSCs and Tregs effectively prevents premature/physiological aging of HSCs.

Summary: This study investigates the susceptibility of Tconvs to Treg suppression during tolerance induction in transplantation. The findings suggest that transplantation tolerance is associated with an increased sensitivity of alloreactive Tconvs to Treg suppression, which is regulated by the expression of the transcription factor Satb1. Targeting Satb1 could potentially be a strategy to improve transplant outcomes by modulating Treg-sparing immunosuppressive therapies.

Summary: This study demonstrates that adoptive transfer of CD8+ T cells can significantly reduce viral load in a T-cell-deficient mouse model of Zika virus (ZIKV) infection. The study also shows that ZIKV infection can induce CD8+ T cell-mediated skin inflammation. These findings suggest a potential role for CD8+ T cell-associated pathologies in patients with CD4+ T cell immunodeficiency after ZIKV infection.
