Summary: In order to ensure the sterility and safety of hematopoietic stem cell (HSC) products, it is important for each hospital to implement and maintain good practice guidelines and regulations for the HSC collection and processing unit. Retrospective analysis of microbial culture results of 11,743 HSC products over a 25-year period revealed a decrease in contamination rate from 0.5% in the first period to 0.2% in the second period. The study suggests that handling HSC products by trained personnel and adopting established protocols, including quality assurance programs, can help decrease the risk of contamination.

Summary: Hematopoietic stem cell transplantation is an effective regenerative therapy for various diseases, but our understanding of the reconstitution process is limited. This study reveals the dynamics of transplanted hematopoietic stem and progenitor cells (HSPCs) in humans. Transcriptomic analysis shows that immunoregulatory neutrophil progenitors expressing high levels of the S100A gene family are enriched in peripheral blood stem cells. Patients with acute graft-versus-host disease (aGVHD) have fewer S100Ahigh immunoregulatory neutrophil progenitors, suggesting a potential criterion for identifying high-risk patients.

Summary: Interaction with microenvironmental factors is crucial for the regulation of hematopoietic stem cell function. In this study, it was found that integrin signaling regulates the expression of SDF-1 alpha through transcriptional regulation. Inhibition of SRC gene or pharmacological inhibition resulted in increased expression of the chemokine SDF-1 alpha. Systemic inhibition of SRC led to increased SDF-1 alpha levels in BM plasma, which improved the homing efficiency of transplanted HSCs and enhanced hematopoietic function.

Summary: Patients with hematological malignancy infected with SARS-CoV-2 have an increased risk of severe disease and mortality. Active hematological disease, lower life expectancy related to primary hematological disease, neutropenia at diagnosis of SARS-CoV-2, ICU admission, and first-line therapy used for coronavirus disease-2019 treatment were found to be related to higher mortality rates.

Summary: The benefit of pre-transplant consolidation treatments remains unclear in patients with acute lymphoblastic leukemia (ALL) undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT). In this study, receiving >= 2 consolidation treatments was associated with better overall survival. Consolidation treatments reduced non-relapse mortality and improved survival.

Summary: This study examined the occurrence of fatigue, insomnia, depression, anxiety, and stress symptoms in patients after allogeneic hematopoietic stem cell transplantation (AHSCT) and explored potential relationships among these symptoms. The results showed that fatigue was the most common symptom, followed by insomnia, depression, anxiety, and stress. There were moderate relationships between these symptoms, with insomnia being strongly associated with fatigue.
