Metabolic rejuvenation upgrades circulatory functions of red blood cells stored under blood bank conditions

Mickael Marin, Camille Roussel, Michael Dussiot, Papa A. Ndour, Olivier Hermine, Yves Colin, Alan Gray, Matt Landrigan, Caroline Le Van Kim, Pierre A. Buffet, Pascal Amireault

Summary: Rejuvenation of red blood cells restores intracellular ATP levels, reduces phosphatidylserine (PS) exposure, and decreases the proportion of morphologically altered red blood cells accumulated during storage. Rejuvenation also improves the filterability of red blood cells and prevents their adhesion to endothelial cells.

Systematic analysis of direct antiglobulin test results in post-artesunate delayed haemolysis

Tommaso Ascoli Bartoli, Luciana Lepore, Alessandra D'Abramo, Giovanna Adamo, Angela Corpolongo, Laura Scorzolini, Maria Letizia Giancola, Nazario Bevilacqua, Claudia Palazzolo, Andrea Mariano, Giuseppe Ippolito, Pierre Buffet, Emanuele Nicastri

Summary: In this study, nearly half of PADH patients had a positive DAT. The presence of a positive DAT may not necessarily indicate a pathogenic role of anti-erythrocyte antibodies in PADH patients, but rather a marker of immune activation.

How the spleen reshapes and retains young and old red blood cells: A computational investigation

He Li, Zixiang Leonardo Liu, Lu Lu, Pierre Buffet, George Em Karniadakis

Summary: The spleen plays crucial roles in immune functions and the red blood cell life cycle. Computational simulations reveal specific mechanisms in the spleen that facilitate the maturation of young red blood cells and clearance of senescent ones. This study provides insights into the physiological role of the spleen and could potentially inform the optimization of in vitro red blood cell culture techniques.

Naive and memory CD4+ T cell subsets can contribute to the generation of human Tfh cells

Raphael Jeger-Madiot, Romain Vaineau, Maud Heredia, Nicolas Tchitchek, Lisa Bertrand, Mathias Pereira, Oceane Konza, Bruno Gouritin, Benedicte Hoareau-Coudert, Aurelien Corneau, Catherine Blanc, Eric Savier, Pierre Buffet, Adrien Six, David Klatzmann, Arnaud Moris, Stephanie Graff-Dubois

Summary: CD4(+) T follicular helper cells play a crucial role in B cell maturation and antibody production. This study investigates the dynamics of naive and memory CD4(+) T cells in the generation of Tfh cell response, and reveals that both types of cells can acquire Tfh cell characteristics. The expression of pro-Tfh genes supports the transition of memory and naive CD4(+) T cells into the Tfh cell profile. HIV infection differentially affects these pathways of Tfh cell generation, providing a new framework for understanding Tfh cell responses.

Altered Subpopulations of Red Blood Cells and Post-treatment Anemia in Malaria

Charlotte Chambrion, Mallorie Depond, Lucia Angella, Oussama Mouri, Eric Kendjo, Aurelie Fricot-Monsinjon, Camille Roussel, Sylvestre Biligui, Ilhame Tantaoui, Aida Taieb, Nicolas Argy, Sandrine Houze, Renaud Piarroux, Jean-Yves Siriez, Stephane Jaureguiberry, Sebastien Larreche, Marc Thellier, Nicolas Cenac, Pierre Buffet, Papa Alioune Ndour

Summary: In acute malaria, the loss of erythrocytes primarily occurs after therapy, leading to anemia. The mechanisms behind this post-treatment anemia are still unknown. This study investigated the changes in RBC subpopulations during severe malaria treated with artemisinin derivatives. The findings showed decreased deformability of RBC, and the loss of RBC was mainly attributed to the elimination of uninfected RBC.

Immune recovery-related patterns of post kala-azar dermal and ocular leishmaniasis in people living with HIV

Stella Rousset, Mathilde Zenou, Aurelie Saunier, Fanny Varenne, Vincent Soler, Emilie Tournier, Lea Legrand, Laurence Lachaud, Pierre Buffet, Antoine Berry, Pierre Delobel, Guillaume Martin-Blondel

Summary: This study reported cases of post kala-azar dermal leishmaniasis (PKDL) in people living with HIV (PLHIV) and compared the characteristics of two groups based on whether PKDL occurred in the context of immune recovery under antiretroviral therapy (ART) or not. The study found that PKDL can occur as part of an immune restoration disease under ART or as a relapse of visceral leishmaniasis in the context of AIDS. Comparisons between the two groups showed that patients in the immune recovery group had higher CD4(+) cell counts, lower rates of concomitant visceral leishmaniasis, and a trend towards more frequent ocular involvement.

Splenic clearance of rigid erythrocytes as an inherited mechanism for splenomegaly and natural resistance to malaria

Benoit Henry, Geoffroy Volle, Hilaire Akpovi, Laure Gineau, Camille Roussel, Papa Alioune Ndour, Felicien Tossou, Felipe Suarez, Friso Palstra, Aurelie Fricot, Charlotte Chambrion, Julien Solinc, Julie Nguyen, Mathilde Gare, Florentin Aussenac, Charles-Henry Cottart, Christine Keyser, Rafiou Adamou, Magali Tichit, David Hardy, Nadine Fievet, Jerome Clain, Andre Garcia, David Courtin, Olivier Hermine, Audrey Sabbagh, Pierre Buffet

Summary: The Fulani ethnic group has a higher deformability of circulating erythrocytes compared to non-Fulani, indicating enhanced clearance of rigid erythrocytes by the spleen. This phenotype is observed in individuals with markers of Plasmodium falciparum infection. The heritability of this trait is high with a strong multigenic component, and genes expressed in the spleen may be involved in the splenic clearance of erythrocytes.

Susceptibility to malaria in fulani, Bariba, Otamari and gando individuals living in sympatry in Benin: Role of opsonizing antibodies to Plasmodium falciparum merozoites

Abdou Khadre Dit Jadir Fall, Ikhlaq Hussain Kana, Asier Garcia-Senosiain, Benoit Henry, Celia Dechavanne, Andre Garcia, Pierre Buffet, Audrey Sabbagh, Florence Migot-Nabias, Michael Theisen, David Courtin

Summary: This study explores a potential mechanism of natural protection against malaria in the Fulani population, suggesting that antibody-mediated merozoite phagocytosis may play a role. Results show that adults from the Bariba, Otamari, and Gando groups have higher levels of opsonic phagocytosis compared to children, while the Fulani population shows the opposite trend. Fulani children have higher levels of opsonic phagocytosis and anti-GLURP antibodies compared to non-Fulani children, with no significant difference observed in Fulani adults. Higher levels of opsonic phagocytosis before the malaria transmission season are associated with negative malaria Rapid Diagnostic Test results after the season.

Marginal-Zone Lymphomas

Claude Matuchansky

Summary: This letter addresses a concern regarding the statement made by the authors in an article that there are no diagnostic biomarkers for marginal-zone lymphomas (MZLs). The author highlights the existence of a diagnostic biomarker, the alpha-CD protein, specifically for immunoproliferative small intestinal disease (IPSID), a form of MZL previously known as alpha-heavy chain disease or Mediterranean lymphoma.
