Summary: The study focused on investigating miRNA biomarkers for KCZ-induced liver injury by integrating in vivo, in vitro, and bioinformatics analyses. Significant dysregulation of miR-34a-5p, miR-331-3p, and miR-15b-3p was associated with cytoplasmic vacuolization in rat livers with KCZ-induced injury. These miRNAs, along with miR-676, showed evolutionary conservation and could potentially serve as translational biomarkers for early detection of liver injury.

Mechanistic validation in neuronal cells confirmed several biologically efficacious drug combinations for reducing oxidative stress and maintaining neuronal viability and morphology. This pipeline highlights the importance of harmonizing heterogeneous and complementary data/knowledge for drug development of complex diseases.

Summary: Biologically important metals impact cellular homeostasis, and anthropogenic exposure to these metals can lead to adverse effects in humans, including increased incidence of diseases. This study aimed to explore the effects of metals and common genes/signaling pathways involved in metal toxicity through toxicogenomic data mining. The study identified common genes, enriched pathways, and identified heart failure as a major disease associated with metal exposure. Exposure to essential metals may lead to adverse effects through inflammation and oxidative stress.
