Evaluation of a urinary kidney biomarker panel in rat models of acute and subchronic nephrotoxicity

Several novel urinary kidney biomarkers were recently approved by the US-FDA and EMA for improved detection of nephrotoxicity, but few data regarding their performance are publicly available so far. In this study, we investigated the potential of some of the newly accepted makers (Kim-1, beta-2-microglobulin, cystatin C, clusterin) along with six additional urinary key proteins of kidney injury (GST-alpha, Timp-1. VEGF, calbindin, NGAL/lipocalin-2, osteopontin) to detect proximal tubule damage in the rat model studying either acute drug-induced kidney injury or subchronic nephrotoxicity. Candidate proteins were measured in urine samples obtained from rats treated with gentamicin (0,60 and 120 mg/kg bw for 7 days), BI-3 [3-pyrrolidineacetic acid, 5-[[(4'-[imino[(methoxycarbonyl) amino]methyl] [1,1'-biphenyl]-4-yl]oxy]methyl]-2-oxo-, methyl ester,(3S-trans)] (0, 100, and 1000 mg/kg bw for up to 14 days) or with the mycotoxin ochratoxin A (OTA) (0, 21, 70 and 210 mu g/kg bw for up to 90 days) using a Luminex (R) xMAP (R) platform. Cystatin C and NGAL appeared to be the most sensitive indicators of gentamicin nephrotoxicity, with significant changes occurring as early as day 1, and importantly before alterations in serum creatinine or blood urea nitrogen (BUN). Altered urinary excretion of KIM-1, clusterin, calbindin and Timp-1 accompanied by a rise in BUN was observed in rats with BI-3 at 1000 mg/kg bw for 14 days. In contrast, histopathological alterations induced by OTA, which preceded effects on traditional clinical parameters, were best reflected by changes in urinary Kim-1. Overall, our data confirm increased sensitivity of new markers as compared to traditional clinical chemistry parameters. (C) 2010 Elsevier Ireland Ltd. All rights reserved.

Summary: Since the 1940s, there have been ongoing discussions about the benefits and risks of adding fluoride to drinking water. A recent review focused on assessing the risk of current exposure levels in Europe to human health. Despite inconsistent data, the evidence does not support the idea that fluoride should be considered a human developmental neurotoxicant at current exposure levels in European countries.

Exploration of zebrafish larvae as an alternative whole-animal model for nephrotoxicity testing

Summary: Zebrafish larvae are considered a promising alternative whole-animal model for assessing systemic toxicity. Exposure of zebrafish larvae to various nephrotoxins resulted in kidney injury and impaired function. Gene expression analysis showed the most prominent changes in larvae treated with gentamicin compared to other nephrotoxins.

In Vitro and In Vivo Analysis of Ochratoxin A-Derived Glucuronides and Mercapturic Acids as Biomarkers of Exposure

Summary: The study found that OTA-derived glucuronides in urine are suitable biomarkers for monitoring, but there is insufficient evidence to support the use of OTA-derived mercapturic acids as biomarkers. There is a linear relationship between urinary glucuronide excretion and OTA exposure, but more data is needed to establish this relationship between metabolites in urine and external dose.

Zebrafish Embryos and Larvae as Alternative Animal Models for Toxicity Testing

Summary: Before conducting experiments involving living animals in biological laboratories, it is important to consider the necessity, ethics, and potential harm. The development of the 3R principle has become a key standard in animal experimentation worldwide, leading to the exploration of alternative methods in species like zebrafish to reduce experiments on mammals. Zebrafish, with their favorable biological properties, have shown promise in toxicological testing, particularly in cardio-, neuro, hepato- and nephrotoxicity studies.

Assessment of aromatic amides in printed food contact materials: analysis of potential cleavage to primary aromatic amines during simulated passage through the gastrointestinal tract

Summary: Recent analyses conducted by German official food control reported the detection of aromatic amides in cold water extracts from certain food contact materials made from paper or cardboard. The experiments using in vitro digestion models showed that these aromatic amides could be cleaved to potentially genotoxic aromatic amines during the simulated passage through the gastrointestinal tract. However, the rate of cleavage was found to be very low, suggesting that the health risks associated with the presence of aromatic amides in food contact materials might be overestimated.

Species-differences in the in vitro biotransformation of trifluoroethene (HFO-1123)

Summary: Inhalation exposure to HFO-1123 resulted in severe toxicity in minipigs and rabbits, but not in rats. This study aimed to evaluate species differences in the metabolism of HFO-1123 and found that minipigs had higher beta-lyase activity, which led to the formation of a highly toxic compound compared to humans. This suggests that humans may be less susceptible to HFO-1123 toxicity compared to minipigs.
