Optimization of diagnostic methods and criteria of endometritis for various postpartum days to evaluate infertility in dairy cows

Postpartum endometritis is the main cause of infertility in dairy cows, but there is a lack of critical diagnostic criteria. We aimed to 1) determine the optimal diagnostic method and criteria of endometritis for various postpartum days to evaluate infertility, and 2) assess the diagnostic accuracy of a combination of diagnostic methods. Holstein dairy cows (n = 441) from nine commercial dairy herds were examined at 42 +/- 7 days postpartum by using 5 methods: 1) transrectal palpation measurement of the cervical diameter, 2) ultrasonographic measurement of the fluid in uterus (FIU) score, 3) vaginoscopic detection of external uterine orifice hyperemia, 4) vaginal discharge score (VDS), and 5) endometrial cytological percentage of polymorphonuclear leukocytes (PMN%). The clinical findings that were significant in the Pearson chisquare test and had the highest sum of sensitivity and specificity for infertility at 100, 125, 150, 175, and 200 days after parturition were determined the optimal criteria of endometritis. Logistic regression and the Cox proportional hazards model were used to compare the accuracies of the different diagnostic methods for infertility at various postpartum days. The combinations of methods which were significant in Pearson chisquare test and had the highest sum of sensitivity and specificity for infertility were proposed as the optimal combination for determination of endometritis status for various post-partum day. The optimal diagnostic criteria were PMN% >= 6.0, FIU >= 2 (continuous hyperechogenic or large amount of storage material), or VDS >= 2 (mucopurulent or worse vaginal discharge) for postpartum days 100 and 125; PMN% >= 8.0, FIU = 3 (large amount of storage material), VDS >= 2, or external uterine orifice hyperemia for day 150; PMN% >= 6.0, FIU = 3, VDS >= 2, or external uterine orifice hyperemia for day 175; and PMN% >= 5.0, FIU = 3, VDS >= 2, or external uterine orifice hyperemia for day 200. Only the results of endometrial cytology were related to infertility regardless of the postpartum days to evaluate infertility or statistical models. Compared with the sensitivity and specificity of a single diagnostic method, the sensitivity of a combination of methods improved but specificity decreased. We concluded that different diagnostic methods and criteria were required for postpartum days to evaluate infertility and diagnostic accuracy was improved by a combination of diagnostic methods rather than by a single method. Elevated Serum Leptin Levels in Patients With Eosinophilic Chronic Rhinosinusitis

Summary: This study found that serum leptin levels are elevated in ECRS patients and are correlated with the proportion of eosinophils in peripheral blood, the number of eosinophils in nasal polyps, and eotaxin-3 mRNA expression in nasal polyps. Furthermore, the study also found that leptin can enhance eotaxin-3 expression in nasal fibroblasts from nasal polyps in a leptin receptor-dependent manner. These findings suggest a significant role for circulating leptin in the development of eosinophilic inflammation in nasal polyps.

Parity affects the relationship between the insemination-ovulation interval and the conception rate in lactating dairy cows

Summary: This study investigated the relationship between parity and the optimal insemination-ovulation interval (IOI) in lactating dairy cows. It was found that ovulation occurring 6-12 hours after artificial insemination (AI) was associated with the highest conception rate in multiparous cows. However, the effect of IOI on the conception rate in primiparous cows was insignificant. Therefore, careful determination of the timing of AI is essential in multiparous cows.

Summary: This study aimed to establish machine learning-based prediction models for nuclear maturation speed (NMS) of Japanese Black beef heifer oocytes using non-invasive indicators. Morphological features were extracted from recorded photos and used to develop the prediction models. The results showed a correlation between predicted NMS and embryo development, suggesting the potential for improving the efficiency of in vitro embryo production.

Pyruvate enhances stallion sperm function in high glucose media improving overall metabolic efficiency

Summary: The presence of a more efficient glycolysis mechanism depending on pyruvate in stallion spermatozoa can counteract the detrimental effects of higher glucose concentrations. Incubating spermatozoa with 10 mM pyruvate in a modified Tyrode's media with 67 mM glucose showed increased motility and maintained viability. This improvement is attributed to the conversion of lactate to pyruvate and increased NAD(+) enhancing glycolysis efficiency.

Differential transcript usage across mammalian oocytes at the germinal vesicle and metaphase II stages

Summary: Ongoing progress in mRNA-Sequencing technologies has contributed to the refinement of assisted reproductive technologies. This study utilized mRNA-Seq datasets of various mammalian species to analyze differential transcript usage (DTU) and identified ten cross-species DTU genes. Gene Ontology (GO) term enrichment analysis revealed the involvement of these genes in RNA and cell-cycle control mechanisms, suggesting their importance during oocyte maturation. Further exploration of these genes' transcript isoforms may lead to the discovery of novel markers for reproductive technologies.

Effects of Nobiletin supplementation on the freezing diluent on porcine sperm cryo-survival and subsequent in vitro embryo development

Summary: This study found that Nobiletin (NOB) can promote the cryosurvival and embryo development of porcine sperm after in vitro fertilization (IVF). The addition of NOB improves sperm motility, acrosome integrity, viability, and mitochondrial membrane potential, reduces lipid peroxidation, and increases cleavage and blastocyst formation rates. Furthermore, NOB treatment upregulates the expression of genes related to pluripotency and cell differentiation in the resulting embryos.

Fluorochloridone induces mitochondrial dysfunction and apoptosis in primary goat Sertoli cells

Summary: This study investigated the cytotoxicity and toxicological mechanisms of fluorochloridone (FLC) on cultured goat Sertoli cells. The results showed that FLC exposure induced oxidative stress, promoted apoptosis and autophagy, inhibited lysosomal biogenesis, and blocked autophagic flux in goat Sertoli cells. FLC also caused mitochondrial dysfunction and disturbed mitophagy, leading to cell apoptosis. The antioxidant N-acetyl-L-cysteine (NAC) reduced FLC-induced ROS accumulation and reversed the disorder of autophagy levels.
