Effect of morphological integrity, period, and type of culture system on the in vitro development of isolated caprine preantral follicles

The aims of this study were the following: (1) to define an optimal period for the IVC of isolated caprine preantral follicles, (2) to verify the relationship between follicular morphology (intact, extruded, and degenerate follicles) and estradiol production, and (3) to evaluate the effects of the bidimensional (2D) and three-dimensional (3D) culture systems on the in vitro development of caprine preantral follicles. Three experiments were performed. In experiments 1 and 2, the isolated secondary follicles were cultured for 18, 24, and 30 days or 30, 36, and 42 days, respectively. In experiment 3, the optimal culture period from experiment 2 was used for 2D and 3D culture systems. After culture, the oocytes were submitted to IVM. The morphological integrity, antral cavity formation rates, follicular diameter, presence of healthy, grown oocytes (>= 110 mu m), rates of resumption of meiosis, and estradiol concentrations were evaluated. In experiment 1, the percentage of oocytes that resumed meiosis was higher in oocytes cultured for 30 days (48.84%) than in oocytes cultured for 18 and 24 days (15% and 20.93%, respectively). In experiment 2, the percentage of oocytes that resumed meiosis was significantly higher in oocytes cultured for 30 and 36 days (47.5% and 50%, respectively) than in oocytes cultured for 42 days (20%). The estradiol concentrations on Day 12 of culture were similar for normal and extruded follicles and higher than those observed in degenerate follicles at the end of the culture period. In conclusion, the 36-day culture period resulted in the highest rates of meiosis resumption. In addition, because the loss of follicular integrity affects the patterns of estradiol production, follicular integrity is a good predictor of follicular quality. (C) 2014 Elsevier Inc. Summary: The type and period of fixation affected the percentage of morphologically normal ovarian follicles. Paraformaldehyde was not recommended as a fixative, while formalin was suitable for 12-hour fixation and Carnoy's solution was efficient for 24-hour fixation. Davidson's solution was the most suitable fixative for evaluating the morphology of ovarian preantral follicles, regardless of the fixation period.

Summary: This study aimed to investigate the relative abundances of ABC transporters mRNA transcripts in ovine ovarian tissue after vitrification and in vitro culture. The results showed a negative correlation between degenerated follicles and mRNA transcript abundances of ABCB1 and ABCG2, with a significant decrease in the percentage of growing follicles after in vitro culture. Further studies are needed to explore the functions of ABC transporters in ovine folliculogenesis after in vitro culture.

How in vitro maturation changes the proteome of ovine cumulus-oocyte complexes?

Summary: The present study evaluated the effects of in vitro maturation (IVM) on the proteome of cumulus-oocyte complexes (COCs) from ewes. The results showed that IVM caused upregulation and downregulation of proteins in COCs. Upregulated proteins in mature COCs after IVM were related to immune response, complement cascade, cell cycle, and extracellular matrix organization, while downregulated proteins in immature COCs after IVM were linked to metabolic processes, immune response, and complement cascade.

Oocyte in vitro maturation with eugenol improves the medium antioxidant capacity and total cell number per blastocyst

Summary: This study investigates the effects of eugenol supplementation on bovine oocyte in vitro maturation and the subsequent in vitro embryo production and quality through IVF. The results show that adding 40μM eugenol to the maturation medium improves antioxidant capacity and cleavage rate. Additionally, lower calreticulin levels and higher total cell numbers in expanded blastocysts were observed in the lower eugenol concentration treatments. Based on these findings, enriching the maturation medium with 10μM eugenol is recommended for in vitro bovine embryo production.

Summary: The study evaluated the proteome of early antral follicles from Ovis aries using LC-MS/MS analysis. A total of 2503 follicle proteins were identified, including abundant proteins such as vimentin, actin, lamin, heat shock proteins, and histones. miRNAs were found to modulate genes involved in cell cycle, cell differentiation, aging, apoptosis, adipocyte differentiation, and cell division in sheep follicles. This comprehensive atlas of proteins expressed in sheep early antral follicles will contribute to the identification of biomarkers for follicular development and oocyte maturation.

Pyruvate enhances stallion sperm function in high glucose media improving overall metabolic efficiency

Summary: The presence of a more efficient glycolysis mechanism depending on pyruvate in stallion spermatozoa can counteract the detrimental effects of higher glucose concentrations. Incubating spermatozoa with 10 mM pyruvate in a modified Tyrode's media with 67 mM glucose showed increased motility and maintained viability. This improvement is attributed to the conversion of lactate to pyruvate and increased NAD(+) enhancing glycolysis efficiency.

Differential transcript usage across mammalian oocytes at the germinal vesicle and metaphase II stages

Summary: Ongoing progress in mRNA-Sequencing technologies has contributed to the refinement of assisted reproductive technologies. This study utilized mRNA-Seq datasets of various mammalian species to analyze differential transcript usage (DTU) and identified ten cross-species DTU genes. Gene Ontology (GO) term enrichment analysis revealed the involvement of these genes in RNA and cell-cycle control mechanisms, suggesting their importance during oocyte maturation. Further exploration of these genes' transcript isoforms may lead to the discovery of novel markers for reproductive technologies.

Effects of Nobiletin supplementation on the freezing diluent on porcine sperm cryo-survival and subsequent in vitro embryo development

Summary: This study found that Nobiletin (NOB) can promote the cryosurvival and embryo development of porcine sperm after in vitro fertilization (IVF). The addition of NOB improves sperm motility, acrosome integrity, viability, and mitochondrial membrane potential, reduces lipid peroxidation, and increases cleavage and blastocyst formation rates. Furthermore, NOB treatment upregulates the expression of genes related to pluripotency and cell differentiation in the resulting embryos.

Fluorochloridone induces mitochondrial dysfunction and apoptosis in primary goat Sertoli cells

Summary: This study investigated the cytotoxicity and toxicological mechanisms of fluorochloridone (FLC) on cultured goat Sertoli cells. The results showed that FLC exposure induced oxidative stress, promoted apoptosis and autophagy, inhibited lysosomal biogenesis, and blocked autophagic flux in goat Sertoli cells. FLC also caused mitochondrial dysfunction and disturbed mitophagy, leading to cell apoptosis. The antioxidant N-acetyl-L-cysteine (NAC) reduced FLC-induced ROS accumulation and reversed the disorder of autophagy levels.

Inhibition of phospholipase C reduces the capacitation of cryopreserved ovine sperm
