Summary: In the simulation prediction of drug-induced ventricular arrhythmia, precomputed simulation result matrices can be used to reduce computation time and improve efficiency in predicting biomarkers such as action potential duration (APD90). Machine learning methods are applied to predict simulation results, leading to a decrease in computation time. This approach can be applied in other fields to reduce computational cost.

Summary: This study found that older age, end-stage renal disease, underlying cardiovascular disease, potential virus mediated cardiac injury, and drugs like hydroxychloroquine/azithromycin contribute to QTc prolongation in COVID-19 patients. The role of ACE inhibitors in preventing QTc prolongation in COVID-19 patients needs further study. Ventricular arrhythmias and mortality were not associated with QTc prolongation in COVID-19 patients.

Summary: Quetiapine is a medication used to treat various mental disorders, but overdose can be life-threatening. A case study of a 75-year-old male who attempted suicide by ingesting a large amount of Quetiapine without other medications was presented, and a complete pharmacokinetic analysis was conducted during his management in the emergency department.

Summary: Targeted therapy and immunotherapy have revolutionized cancer treatment, but oral toxicities induced by these therapies remain a concern. These toxicities can manifest as mucosal lesions, salivary gland dysfunction, or orofacial neuropathies, impacting patients' quality of life and clinical outcomes. Further research is needed to better understand and manage these adverse events.

Summary: This study provides a comprehensive review of the safety profile of biotherapy and molecular targeted therapies in NEN patients, focusing on cardiovascular toxicities. Results indicate that SSAs and TPH inhibitors are safer in terms of overall toxicity, particularly cardiovascular toxicities, compared to mTOR inhibitors and TKIs.

Summary: This systematic scoping review compares the toxicities experienced by patients receiving immune checkpoint inhibitors (ICIs) or targeted therapy (TT) for stage III and IV melanoma. The results show that ICIs have lower rates of adverse events (AEs) compared to TT, but they are associated with higher rates of long-term or permanent AEs.

Summary: Tyrosine kinase inhibitors (TKIs) have greatly improved the treatment of chronic myeloid leukemia (CML), but they can cause serious cardiopulmonary toxicities, such as vascular adverse events, QT prolongation, heart failure, pleural effusion, and pulmonary arterial hypertension. Currently, there are no dedicated clinical management guidelines for TKI-induced toxicities. This review aims to discuss these cardiopulmonary toxicities and proposes a practical guide for their management.

Summary: This retrospective multicenter cohort study evaluated patients who received BRAF/MEK inhibitors and had a progression-free survival of 4 years or more, and found that over 75% of patients still had durable antitumor responses after nearly 8 years of median follow-up. Similar results were observed in patients who discontinued therapy. Long-term toxicities were uncommon and low-grade.

Summary: In this study, the potential of RGD1-R6 peptide carrier as a non-viral vehicle for targeted siRNA delivery to endothelial cells was evaluated. The results showed that the modified siRNA complexes had higher gene silencing efficiency and lower endothelial cell migration compared to unmodified complexes. In a rat endometriosis model, the combination therapy of the carrier with anti-VEGF siRNA complexes significantly inhibited implant growth and VEGFA gene expression, comparable to Dienogest.

Summary: The QTc/JTc interval is significantly and independently prolonged after sudden cardiac arrest, regardless of known QT prolonging triggers. Normalization of the QTc is expected only after day 6 of hospitalization. Assessment of the QTc for determining the etiology of arrest or monitoring the effect of QT prolonging medications may be unreliable.

Summary: A study conducted in 2017 in France found that 36% of heart failure patients were smartphone users, and these users tended to be younger, more frequently male, have a higher educational level, live in cities, and be professionally active. Smartphone users were also less likely to have diabetes, hypertension, atrial fibrillation, or ischemic cardiopathy, but only 22% of patients were actively participating in therapeutic education programs.

Summary: This study investigated the profile and outcome of elderly patients hospitalized for Acute Heart Failure (AHF) in cardiogeriatrics departments in Val-de-Marne area of Paris. Patients in cardiogeriatrics departments were older, less independent, more often depressed, and had a higher Human Development Index compared to those in cardiology departments, but had similar prognosis in terms of AHF rehospitalization at 90 days.

Summary: Cardiac amyloidosis is a significant risk factor for heart failure patients, leading to higher death and hospital readmission rates compared to those without this condition. This study found that patients with cardiac amyloidosis had elevated heart rate and brain natriuretic peptide levels, and lower blood pressure, weight, and body mass index values upon discharge. Furthermore, cardiac amyloidosis patients experienced higher death and hospital readmission rates, occurring sooner after discharge, highlighting the importance of active management for these patients.
