A new approach for the synthesis of multi-functionalized 1,5-benzodiazepines is developed starting from o-phenylenediamines, -keto esters and aromatic aldehydes utilizing a one-pot, three-component strategy employing bromodimethylsulfonium bromide as the catalyst. The simple reaction procedure, good yields, mild reaction conditions and applicability to a wide range of substrates are some of the salient features of this protocol.

Sidechain-Backbone Tetrel Bonding Interactions Provide a General Mechanism for trans-Peptoid Stabilization

Summary: From X-ray, NMR, and DFT studies of azapeptoids, a tetrel bonding interaction that stabilizes trans-peptoids has been discovered. Peptoids with alpha-heteroatoms and N-aryl groups in the sidechain adopt trans-amide geometries due to the tetrel bonding interaction between the sidechain alpha-heteroatom lone pair or pi-electrons and the sigma* orbital of the backbone C-alpha-N bond. CD spectroscopic studies of oligo-proline host-guest model peptides also showed that azapeptoid residues stabilize polyproline II helical conformation.

Synergistic n → π* and nN → π*Ar interactions in C-terminal modified prolines: effect on Xaa-Pro cis/trans equilibrium

Summary: Carbonyl-carbonyl (COMIDLINE HORIZONTAL ELLIPSISCO) n -> pi* interaction often coexists with a hydrogen bond or another n -> pi* interaction. In this study, a set of proline-diacylhydrazine molecules were investigated, and it was found that the increase in the strength of the n(N) -> pi*(Ar) interaction on the DAH side strengthened the n -> pi* interaction on the Pro side. This research provides a simple C-terminal modification strategy to stabilize the trans-Pro geometry, which is important for stabilizing PPII helices and collagen triple helices.
