Summary: Studies have found that inhibitors of matrix metalloproteinase (MMPs) as anti-tumoral drugs have been disappointing. Recent research shows that endothelial cells and endothelial colony forming cells (ECFCs) can switch invasion modes and perform amoeboid angiogenesis in the absence of protease activity. TIMP1, TIMP2, and TIMP3 have been identified as high-powered protease inhibitors that can induce amoeboid invasion of ECFCs, while Marimastat promotes ECFC invasion and tubular formation. It is important to consider the amoeboid mechanism of invasion in developing new cancer drugs.

Summary: Angiogenesis is a crucial process for the growth and metastasis of gastric cancer. Angiopoietin-2 is an important driver of tumor angiogenesis and has shown potential as a target for antiangiogenic therapy. Extracellular vesicles play a significant role in tumor progression, including angiogenesis. This study investigated the interaction between gastric cancer and endothelial cells mediated by vesicles, focusing on angiopoietin-2. The results demonstrated the expression of angiopoietin-2 in primary gastric cancer tissues and omental metastasis tissues. Gastric cancer vesicles were found to induce proliferation, migration, invasion, and tube formation of endothelial cells. The vesicles also contained high levels of proangiogenic proteins, including angiopoietin-2. Knockdown of angiopoietin-2 showed that it mediates the proangiogenic effects of gastric cancer vesicles. These findings reveal a novel mechanism by which gastric cancer cells induce angiogenesis and suggest its potential as a target for cancer therapy.

Summary: Colorectal cancer is a common disease and cause of death worldwide. The drug used for second-line treatment depends on the biomarker status and the drugs used in first-line treatment. Several biomarkers have been evaluated in clinical practice, and corresponding targeted therapies are selected based on biomarker status. If all biomarkers are negative, an angiogenesis inhibitor is often used as second-line treatment. Although no useful biomarkers have been established for selecting angiogenesis inhibitors, previous studies suggest that angiogenesis-related factors like VEGF-A and VEGF-D may be predictors of their efficacy. The rationale for selecting these three angiogenesis inhibitors in second-line treatment should be clarified.

Summary: In this study, it was found that MCP-1 promotes VEGF production while miR-374b-5p inhibits it by targeting the 3'-untranslated region of VEGF mRNA. MCP-1 also enhances the acetylation of transcription factor stat3, resulting in decreased expression of miR-374b-5p and increased VEGF production. Inhibiting stat3 acetylation, DNMT1, or EZH2 can effectively inhibit MCP-1-induced neovascularization and tumor growth.
