Summary: In this study, single-cell transcriptomic analysis was used to identify new biomarkers for nucleus pulposus (NP) and inner annulus fibrosis (iAF) cells in intervertebral discs (IVD). The analysis also provided insights into the cell populations within non-degenerating and degenerating IVD. The findings suggest potential functional differences linked to degeneration and among NP and iAF subpopulations, and also propose specific cell type and predictive IVD degeneration genes.

Summary: This study aimed to determine if combining o-Vanillin and RG-7112 could more efficiently remove senescent cells and reduce inflammatory factors and pain mediators in degenerating intervertebral disc (IVD) cells. The results showed that the combination of these two senolytic drugs significantly reduced the amount of senescent IVD cells, proinflammatory cytokines, and neurotrophic factors. These findings support the potential of senolytics as a promising treatment for IVD-related low back pain.

Summary: MSCs are being tested as a cell therapy in clinical trials for inflammatory disorders, but suitable preclinical animal models are rare. In this study, two robust animal models of immune pathology were introduced to study the effect of IFN gamma primed bone marrow derived MSCs in acute radiation syndrome and graft versus host disease. The results showed that infused IFN gamma primed allogeneic MSCs protect against lethality from ARS, but not GvHD.

Summary: The study demonstrated that patients with intervertebral disc degeneration had increased levels of fibrotic markers in their tissues. Bleomycin induced a pro-fibrotic effect on annulus fibrosus cells and bone marrow stromal cells through the TGF beta-TGF beta R1-SMAD2/3 signaling pathway. Pharmacological inhibition or gene knock-down of TGF beta R1 could mitigate the pro-fibrotic effects.

Summary: This study assessed the association between lumbar bone marrow adipose tissue fat fraction (BMAT-FF) and paraspinal muscle proton density fat fraction (PDFF) with intervertebral disc degeneration (IVDD). The results showed a significant correlation between BMAT-FF and PDFF, and both factors were found to be independent and additive risk factors for IVDD. The findings suggest that quantitative MRI measurements of paraspinal myosteatosis and vertebral bone marrow fatty infiltration can serve as imaging biomarkers to assess the individual risk for IVDD.

Summary: The application of mesenchymal stromal/stem cells (MSCs) and their exosomes in treating degenerative diseases, especially intervertebral disc disease (IDD), shows promising results. MSCs exert therapeutic effects by differentiating, reducing apoptosis, increasing proliferation, and decreasing senescence in nucleus pulposus cells (NPCs). The application of MSCs and their exosomes also affects annulus fibrosus and cartilaginous endplate cells, preventing the progression of disc degeneration.
