Summary: This meta-analysis evaluated the evidence for the use of parathyroid hormone (PTH) analogues to improve fracture healing. PTH improved functional outcomes but did not affect fracture healing rate or reduce pain. The evidence for PTH analogue use in fracture healing is less clear in younger, non-osteoporotic patient populations, warranting further research.

Summary: This study compared the effects of two osteoanabolic drugs, ABL and TPTD, on protecting alveolar bone in experimental periodontitis. The results showed that both ABL and TPTD reduced alveolar bone loss, with ABL having better osteoanabolic effects. In vitro experiments also demonstrated that ABL had higher alkaline phosphatase activity and osteoblastic gene expression compared to TPTD.

Summary: This study confirms the efficacy of teriparatide in improving fusion rates in osteoporotic patients undergoing spinal fusion surgery. Patients treated with teriparatide had significantly higher rates of lumbar spinal fusion compared to those not receiving the treatment, with a greater likelihood of fusion compared to bisphosphonate and placebo cohorts. The treatment also showed benefits in reducing subsequent vertebral fractures, sagittal malalignment, limb VAS, and spinal VAS. Further research is needed to determine the optimal duration of treatment and timing of surgery.

Summary: This study aims to evaluate the effectiveness of teriparatide in improving radiographic and functional outcomes after spinal fusion surgery for osteoporosis. Through systematic review and network meta-analysis, the results showed significant differences in radiological outcomes, with TP group exhibiting better outcomes compared to the control group.

Summary: This study identifies SIK2/SIK3 as potential drug targets for treating osteoporosis and successfully develops an orally available SIK2/SIK3 inhibitor. The inhibitor stimulates bone formation and increases bone density without apparent toxicity. These findings provide a new approach for the pharmacological treatment of osteoporosis.

Summary: This study extracted a dataset of 62 miRNAs from different studies and constructed an interactome of PTH-regulated miRNAs and their target genes. Several miRNAs with significant interactions were identified, providing further understanding of the mechanism of action of PTH in bone metabolism and osteoporosis.

Summary: This review compiled and discussed the mechanisms underlying the effects of parathyroid hormone (PTH) on bone healing following distraction osteogenesis. The findings showed that PTH accelerates bone regeneration through its involvement in cell proliferation and differentiation, bone formation, and callus remodeling.

Summary: The study investigated the serum concentration of various markers in healthy-weaned-foals and found significant correlations between OC, PTH, CT, APL values and foal's age, as well as a positive correlation between PTH levels and OC values in both fillies and colts. Sex-related differences in OC concentration were also observed, indicating a possible involvement of sex hormones in regulating bone metabolism in prepubertal foals.

Summary: The study showed that PTH and Scl-Ab can decrease the expression of miR-19a/b in bones, while antagonizing miR-19a/b can increase bone mass. Furthermore, antagonizing miR-19a/b enhances bone mass gain and restores bone loss by supporting bone formation and decreasing bone resorption.

Summary: This study revealed that mice lacking ProCT showed increased bone resorption and excessive osteoclast formation during iPTH treatment. ProCT inhibits osteoclast formation by arresting macrophage migration, leading to reduced bone resorption during iPTH.

Summary: The study revealed that PTH signaling in mature osteoblasts/osteocytes is crucial for protecting the skeleton from age-induced bone loss by restraining osteoclast activity and protecting osteocytes from oxidative stresses.

Summary: The study demonstrated that preoperative PTH administration significantly reduced the number of empty lacunae and increased the number of blood vessels, suggesting more viable bone tissue in this group. However, postoperative PTH administration showed no significant differences compared to controls. Therefore, preoperative PTH use may represent a better prophylactic regimen for preventing MRONJ.

Summary: Teriparatide, also known as 1-34 parathyroid hormone, is commonly used for treating osteoporosis in postmenopausal women. However, its therapeutic potential is limited by stability issues and low bioavailability. A glycosylated derivative, PTHG2, was designed to improve stability and anti-osteoporosis effects. In animal models and in vitro experiments, PTHG2 promoted bone formation, inhibited marrow adipogenesis, and directed BMSCs fate through the cAMP-PKA signaling pathway.

Summary: Residual ridge resorption is a chronic bone resorption that occurs after tooth loss. This study used a mouse model to analyze long-term changes in bone morphology and gene expression. The findings suggest that hormonal changes and inhibition of specific molecules may be potential strategies for treating residual ridge resorption.
