Summary: Adenosquamous carcinoma of the pancreas (ASCP) is a more aggressive subtype of pancreatic adenocarcinoma compared to pancreatic ductal adenocarcinoma (PDAC), with poorer differentiation and higher rates of lymph node metastases. Despite a shorter median survival, 5-year survival rates after surgical resection of about 18% can be expected in ASCP, supporting resection as part of a multimodal therapy as the treatment of choice in this rare cancer.

Summary: This study utilized single-cell sequencing to investigate pancreatic adenosquamous carcinoma (ASPC) and identified multiple cell types and immune microenvironment differences among IPMN, ASPC, and PDAC. Two novel subtypes of cancer-associated fibroblasts and ASPC cancer cells with squamous cell features were also identified. These findings have important implications for the clinical application of single-cell sequencing data and advancing our understanding of ASPC.

Summary: This study found that PDAC patients with imaging necrosis had a worse prognosis, with shorter recurrence-free survival and overall survival compared to those with neoplastic mucin cysts. Besides, PDAC with imaging necrosis exhibited more histological necrosis, more aggressive tumor differentiation, and higher tumor cellularity than PDAC with neoplastic mucin cysts.

Summary: The tumor microenvironment (TME) is crucial for the development, progression, and metastasis of pancreatic cancer (PC), especially in adenosquamous carcinoma of pancreas (ASCP). The expression of CD3, CD4, CD8, FoxP3, and PD-L1 within the TME was studied, and their correlation with the prognosis of PC was identified. Higher levels of PD-L1 were associated with shorter overall survival in ASCP and PDAC patients, while higher infiltration of CD3+ and CD8+ T-cells was significantly correlated with better prognosis in PC.

Summary: Adenosquamous carcinoma (ASC) of the pancreas is a rare and aggressive malignancy that mainly affects women with a median age of 73 years. Most cases present with tumors in the head of the pancreas, show a wide range of clinical manifestations, and are primarily treated with oncological resection.

Summary: This study identified two reliable immune-related gene signature models that were significantly associated with prognosis and tumor microenvironment cell infiltration in LUAD and LUSC, respectively. Evaluation of these models could help predict response to immunotherapy and guide immunotherapy strategies.

Summary: The composition of pulmonary microbiota in tissues adjacent to lung lesions showed significant differences between different lesion types. Ralstonia, Acinetobacter, and Microbacterium are the dominant genera in lung adenocarcinoma, lung squamous carcinoma, and benign lesions, respectively. The abundance ratio of Ralstonia to Acinetobacter in adenocarcinoma patients has diagnostic value. PICRUSt analysis revealed remarkable differences in metabolic pathways among different lesion types.

Summary: This study retrospectively evaluated 78 patients with gastric-type adenocarcinoma and adenosquamous carcinoma between 2000 and 2020, and identified two cases with gastric-type adenocarcinoma showing squamous differentiation. These tumors were in advanced stage and had a mixture of gastric-type adenocarcinoma and squamous cell carcinoma components without HPV-related features.

Summary: Primary pancreatic squamous cell carcinoma is sporadic and often has a poor prognosis with no established treatment guideline. This case report demonstrates the effectiveness of immunotherapy in combination with chemotherapy against locally advanced squamous cell carcinoma of the pancreas with high PD-L1 expression, resulting in symptom-free survival for over 16 months.

Summary: The expression levels of GPER1 are found to be associated with tumor type and survival rates in esophageal carcinoma. In overall esophageal carcinoma, low expression of GPER1 is correlated with poor survival rates, while overexpression of GPER1 is associated with the development of esophageal adenocarcinoma and its pre-cancerous lesion. Furthermore, the protein expression of GPER1 is higher in esophageal adenocarcinoma tissues compared to normal esophageal tissues, whereas it is lower in esophageal squamous cell carcinoma tissues.

Summary: In this study, Raman spectroscopy and deep learning were utilized for accurate and stain-free intraoperative diagnosis. By treating Raman spectral signals as sequences and converting them into two-dimensional Raman spectrograms using short-time Fourier transform as input, high performance deep learning models were achieved, surpassing traditional methods in accuracy and sensitivity.

Summary: The study assessed the ability of ring-enhancing sign and focal necrosis to diagnose ASqC on MRI and CT. Results showed that MRI displayed ring enhancement in ASqC with higher reader confidence compared to CT and FN in ASqC with greater inter-reader agreement. The concurrent presence of these two imaging features should raise high suspicion for ASqC.

Summary: The study found associations between pre-operative ERCP and ERCP frequency with clinical characteristics and islet yield in patients undergoing TPIAT. However, pre-operative ERCP did not significantly impact islet yield. Therefore, ERCP can be performed as needed without withholding to optimize surgical outcomes.

Summary: The study aimed to identify baseline clinical and imaging metrics to predict the need for intervention in patients with necrotizing pancreatitis. Independent predictors of eventual intervention included white race, transferred status, need for TPN, size of necrotic collection, and extent of encapsulation. A prediction model incorporating these factors showed a high accuracy in predicting the need for intervention.
