Summary: Polyphenolic compound Oleuropein aglycone, found in abundance in olive oil from the Mediterranean diet, has been shown to inhibit amyloid self-assembly of pro-inflammatory protein S100A9 and mitigate its damaging effects on neuroblastoma cells. By interacting directly with native and fibrillar S100A9, Oleuropein aglycone prevents amyloid oligomerization, reduces the length of amyloid fibrils, disintegrates preformed fibrils, and decreases amyloid load. This compound also targets S100A9 amyloids at the cellular level, reducing amyloid accumulation and increasing overall cell viability in the presence of amyloid load.

It did not affect viral protein expression. The peptide also demonstrated anti-inflammatory activity and efficacy against an acyclovir-resistant HSV-2 strain.

Summary: Cyclic dinucleotides (CDNs) are signaling molecules that exist in all living organisms. This study investigates the mechanism by which the solute carrier protein SLC19A1 recognizes and transports CDNs, folate, and antifolate substances. Cryo-electron microscopy structures of SLC19A1 with different CDNs, folate, and antifolate were analyzed, revealing unique recognition and transport mechanisms for each substrate. This research provides insights into the function of SLC19A1 and has implications for the development of potential therapeutics.
