Summary: Enhancer of zeste homolog 2 (EZH2), an overexpressed therapeutic target molecule in various cancers, has immunomodulatory functions in the tumor microenvironment (TME). This study evaluated the impact of EZH2 on TME of hepatocellular carcinoma (HCC) and found that EZH2 inhibitor impaired tumor growth but its effect was weakened in immunocompetent mice. Combining EZH2 inhibitors with agents that reduce MDSCs might represent a novel therapeutic strategy for HCC.

Summary: This study identifies aberrant enhancers and associated transcriptional dysregulation in hepatocellular carcinoma (HCC) using integrative analysis of various DNA sequencing data. The study also explores the functions and clinical significance of these aberrant enhancers. Furthermore, a prognostic signature based on aberrant enhancers is established with excellent predictive accuracy for overall survival of HCC patients.

Summary: miR-3168 is increased in liver cancer stem cells (CSCs) and inhibits CSC expansion by targeting p53 directly. Additionally, miR-3168 may be a predictor of survival benefits from transcatheter arterial chemoembolization (TACE) in hepatocellular carcinoma (HCC) patients. The study uncovers a novel mechanism of liver CSC regulation and provides a potential therapeutic target for liver CSCs.

Summary: RNA-binding protein MBNL2 plays a crucial role in regulating tumor cell proliferation and DNA damage response by modulating the PI3K/AKT pathway and affecting the expression of p21. Depletion of MBNL2 leads to increased phosphorylation levels of Chk1 and influences DNA damage response, ultimately altering tumor cell fate by inhibiting DNA damage repair and promoting DNA damage-induced apoptosis.

Summary: SERPINE2 is identified as a regulator of radiosensitivity and the DNA damage response (DDR) in lung cancer, with knockdown improving tumor radiosensitivity by reducing homologous recombination repair, restoring cell cycle checkpoints, and suppressing migration and invasion. Additionally, high SERPINE2 expression correlates with poor prognosis and a high serum concentration predicts a poor response to radiotherapy in lung cancer patients.

Summary: This study demonstrated the upregulation of TL1A, IFN gamma, and two MLCK variants in human IBD biopsy specimens. TL1A was found to induce MLCK-dependent changes in gut barrier function and bacterial internalization. Different MLCK isoforms were shown to orchestrate divergent pathways of bacterial infiltration in IBD, suggesting a potential mechanism for symptom flares in the disease.

Summary: This study aimed to evaluate the prevalence, clinical manifestation, and outcomes of primary sclerosing cholangitis (PSC) in Taiwanese patients with inflammatory bowel disease (IBD). The results showed a prevalence rate of 1.57% for PSC in Taiwanese IBD patients. The majority of patients with IBD and PSC were men and were diagnosed at a younger age. Routine evaluation of biliary enzymes and liver imaging is recommended in young male patients with IBD.

Summary: This study evaluated the impact of the COVID-19 pandemic on the perceptions of patients with inflammatory bowel disease (IBD). The results showed that patients were concerned about adverse events from vaccination, higher risks of developing severe COVID-19, and COVID-19 infection compared to the general population. Some patients self discontinued IBD medication during the pandemic, leading to worse symptoms. However, after receiving education, the anxiety levels of the patients were mitigated.

Summary: This study investigated the current approaches and practices of physicians regarding vaccinations for inflammatory bowel disease (IBD) patients in different Asian countries/regions. The results showed similarities among countries/regions in terms of recognizing the importance of vaccination, but there were differences in the selection and recommendation of certain vaccines, reflecting each country's vaccination guidelines and health insurance. Therefore, more communication among doctors and a consensus regarding IBD vaccination differences in the Asian region are needed.
