Apoptosis Signal-regulating Kinase 1 promotes Ochratoxin A-induced renal cytotoxicity

Oxidative stress and apoptosis are involved in Ochratoxin A (OTA)-induced renal cytotoxicity. Apoptosis signal-regulating kinase 1 (ASK1) is a Mitogen-Activated Protein Kinase Kinase Kinase (MAPKKK, MAP3K) family member that plays an important role in oxidative stress-induced cell apoptosis. In this study, we performed RNA interference of ASK1 in HEK293 cells and employed an iTRAQ-based quantitative proteomics approach to globally investigate the regulatory mechanism of ASK1 in OTA-induced renal cytotoxicity. Our results showed that ASK1 knockdown alleviated OTA-induced ROS generation and Delta psi m loss and thus desensitized the cells to OTA-induced apoptosis. We identified 33 and 24 differentially expressed proteins upon OTA treatment in scrambled and ASK1 knockdown cells, respectively. Pathway classification and analysis revealed that ASK1 participated in OTA-induced inhibition of mRNA splicing, nucleotide metabolism, the cell cycle, DNA repair, and the activation of lipid metabolism. We concluded that ASK1 plays an essential role in promoting OTA-induced renal cytotoxicity.

Lonp1 and Sig-1R contribute to the counteraction of ursolic acid against ochratoxin A-induced mitochondrial apoptosis

Summary: Previous studies have shown that ursolic acid (UA) can protect against ochratoxin A (OTA)-induced nephrotoxicity through Lonp1. This study investigated whether other proteins located in the mitochondria-associated endoplasmic reticulum membranes (MAMs), such as Sig-1R, also contribute to the protection of UA against OTA-induced nephrotoxicity. The results showed that OTA treatment induced apoptosis in HK-2 cells through the repression of Lonp1 and Sig-1R, and this effect was relieved by pretreatment with UA.

Application of the Reuterin System as Food Preservative or Health-Promoting Agent: A Critical Review

Although the antimicrobial mechanism and toxicity of the reuterin system are still unclear, this updated review provides valuable information on its biosynthesis, composition, production, mechanisms, stability, toxicity, and potential applications. Challenges and opportunities for using the reuterin system as a food preservative or health-promoting agent are also discussed.

Excessive ER-phagy contributes to ochratoxin A-induced apoptosis

In vitro and in silico studies of the structure and functional properties of the lactoferrin-chlorogenic acid complex

Summary: In this study, chlorogenic acid (CGA) was used to interact with lactoferrin (LF) to form nanoscale complex. The structure and function of LF complexed with CGA at different ratios were analyzed. The interaction between LF and CGA changed LF's characteristic UV and fluorescent spectra, and the fluorescence quenching mechanism was static quenching. The LF-CGA complex showed decreased hydrophobicity and increased solubility, and its foaming ability and antioxidant activity in vitro were enhanced.

Characterization and the cholesterol-lowering effect of dietary fiber from fermented black rice (Oryza sativa L.)

Summary: Black rice was fermented using Neurospora crassa to extract dietary fiber (DF), which was then characterized and evaluated for its cholesterol-lowering effect in mice. The findings showed that fermentation increased soluble DF content and enhanced DF's adsorption capacity for various substances. The fermented DF also had a different structure compared to unfermented DF. Additionally, feeding mice with DF from fermented black rice led to weight loss, reduced total cholesterol levels, and improved lipid profile.
