Summary: This study explored the association between RIPK1-dependent necroptosis and the pathogenesis of rheumatoid arthritis (RA), and identified a potential new treatment option. The plasma levels of RIPK1 and MLKL were found to be higher in RA patients and positively correlated with disease severity. Treatment with KW2449, a small molecule inhibitor targeting RIPK1, reduced joint inflammation and tissue damage in an animal model of RA. These findings suggest that RIPK1-dependent necroptosis could be a therapeutic target for RA.

Summary: This study investigated the role of alpha 1 adrenoceptor in the ventrolateral orbital cortex of rats with repeated morphine treatment and its underlying molecular mechanisms. The results showed that prazosin enhanced the addictive effects of morphine and affected neurotransmitter NE and peripheral inflammatory factors. These findings suggest that the NE system may be a potential therapeutic target in morphine addiction.

Summary: Methotrexate (MTX) is commonly used as a first-line therapy for rheumatoid arthritis (RA), but its side effects can limit its use. Folate-targeted liposomal formulation of MTX (FL-MTX) is a modified nanotherapeutic treatment that has shown good therapeutic efficacy in a mouse model of collagen-induced arthritis (CIA). FL-MTX can reduce the hepatic and renal elimination of MTX, leading to a potential reduction in treatment frequency and overall dosage.

Summary: Dantrolene inhibits Ca2+ leakage from destabilized ryanodine receptors and shows potential as a therapeutic agent against endoplasmic reticulum stress-associated diseases. In a mouse model of collagen-induced arthritis, oral administration of dantrolene significantly reduces arthritic scores, suppresses bone and chondral destruction, and inhibits the production of serum anti-type II collagen IgG. These findings suggest that dantrolene may be a potential anti-rheumatic drug.
