Polymorphisms of the serotonin-2A receptor and catechol-O-methyltransferase genes: a study on fibromyalgia susceptibility

Genetic and environmental factors are thought to play roles in the etiopathology of fibromyalgia syndrome (FMS). The objective of this study was to determine the potential effects of single nucleotide polymorphisms (SNPs) in catechol-O-methyltransferase (COMT) (rs4680) and 5-hydroxytryptamine (serotonin) 2A (5-HT2A) receptor (rs6313 and rs6311) genes on susceptibility to FMS. One hundred seventy-one women (80 FMS, 91 control) were enrolled in the study. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used for the genotyping analyses. Genotype and allele frequencies were calculated by the chi-square test. Beck depression inventory, state and trait anxiety inventory and symptom checklist-90 revised (SCL-90-R) tests were applied to both patients and controls. There were no observed differences in the frequencies of alleles and genotypes between patients and controls for the COMT, and the two 5-HT2A receptor gene polymorphisms (P > 0.05). Our results suggest that the investigated polymorphisms seem not to be the susceptibility factors in etiology of FMS.

However, there was no significant correlation between SDF and methylation of XRCC1 and ERCC2 genes.

Summary: The study found that AR CAG repeat expansion might be a risk factor for idiopathic male infertility in Turkish men, while the genotypes of SRD5A2 and TNF-alpha were not significantly associated with the risk of infertility. Further studies are needed to understand the possible associations among AR CAG, V89L, and -308G/A polymorphisms and male infertility.

Summary: The study found that chromatin condensation in sperm of infertile men was significantly lower than that of fertile men, and age, sperm concentration, morphology, and motility were significantly negatively correlated with chromatin condensation. However, no significant correlations were observed among chromatin condensation, SDF, and sperm DNA damage in terms of 8-OHdG concentration.

Summary: In this study, it was found that miR-128/193a-5p/494 may be associated with imatinib resistance in prostate cancer. Specifically, the upregulation of FYN may suppress the transcriptional activity of miR-128/193a-5p/494. Thus, miR-128/193a-5p/494 may play a critical role in imatinib resistance through the tr-KIT pathway.

Summary: This study aimed to characterize the clinical, cytogenetic, and molecular findings of 12 infertile men with isodicentric Y-chromosome abnormalities and found that different breakpoints of Y-chromosome were associated with male infertility. Sperm retrieval outcomes using microTESE showed the possibility of finding sperm in patients with idic(Yp) or idic(Yq).

Among the 105 included studies, food- and water-borne diseases were the most frequently studied. The number of burden of infectious disease studies has increased over time, but there is less consensus in methodological approaches.

Summary: This study provides evidence for the positive effect of varicocele repair on conventional semen parameters in infertile men with clinical varicocele. There is significant improvement in sperm concentration, total motility, progressive motility, total sperm motility, and normal sperm morphology after varicocele repair. However, there is high inter-study heterogeneity.

Semiquantitative promoter methylation of MLH1 and MSH2 genes and their impact on sperm DNA fragmentation and chromatin condensation in infertile men

Summary: The promoter methylation of MLH1 and MSH2 genes may have an impact on sperm DNA packaging and conventional semen parameters, respectively. MLH1 promoter methylation was positively correlated with the percentage of aniline blue positive spermatozoa, while MSH2 promoter methylation was negatively correlated with sperm concentration and total sperm count.
