Summary: This review summarizes recent advances in Takayasu arteritis (TAK), focusing on pathogenesis, imaging modalities, and management. Three novel clusters based on angiographic findings were identified and replicated in different cohorts. New imaging modalities have shown promising results in assessing arterial inflammation. There is more evidence on the long-term use of tocilizumab, but relapses are common. Janus kinase inhibitors seem to be promising based on recent findings. Improvements in imaging modalities and understanding of disease pathogenesis will enhance disease assessment and identification of effective therapies.

Summary: This review aims to provide a comprehensive understanding of the pathophysiology, cause, and epidemiology of Takayasu arteritis, as well as the diagnosis, prognosis, and treatment in children. Although the exact etiology of this disease is not fully understood, it is believed to have an autoimmune basis and genetic predisposition. Unlike in adults, Takayasu arteritis in children has distinct diagnostic criteria and clinical manifestations. Current treatment algorithms involve the use of less toxic contemporary biologic agents and early diagnosis for better long-term prognosis.

Summary: This study demonstrated elevated levels of mast cell activation markers in the sera of TAK patients compared to healthy donors, along with increased mast cell expression in aortic lesions of TAK patients. Mast cells stimulated by TAK patient sera supported neoangiogenesis and fibrosis compared to healthy donor sera, indicating their key role in vascular lesions. These findings suggest mast cells may serve as a potential therapeutic target in large vessel vasculitis.

Summary: Takayasu arteritis (TA) is a rare large vasculitis with unknown etiology that affects the aorta and its primary branches, as well as the pulmonary and coronary arteries. The pathogenesis of TA involves cellular and humoral immunity, chronic inflammation, and genetic factors. Various biomarkers associated with the pathogenesis of TA can be observed in patients, and they have significant clinical application values in early diagnosis, evaluating disease activity, and guiding clinical treatment options. The combination of biomarkers assay and imaging examination may enhance the accuracy of TA detection.

A case of a 15-year-old female initially diagnosed with Crohn's disease and later with Takayasu arteritis is reported. A review of literature showed that in most cases, Crohn's disease was diagnosed first followed by Takayasu arteritis.

Summary: This study aimed to evaluate fertility, early menopause, and pregnancy outcomes of patients with Takayasu's arteritis (TA) before and after diagnosis. The results showed that TA patients had a higher infertility rate compared to the general population in Turkey (12.5% vs 8.6%) and a lower rate of early menopause (16.7% vs 36.1%). Pregnancy following a TA diagnosis had more maternal complications, and fetal complications were common both before and after diagnosis. Chronic inflammation may contribute to increased infertility and fetal complications.

Summary: This study evaluated the efficacy of tocilizumab in patients with refractory Behcet disease. The results showed that 83% of patients responded to tocilizumab, with 60% achieving complete response and 23% achieving partial response. Tocilizumab also significantly reduced steroid dosage and the need for concomitant disease-modifying antirheumatic drugs. However, a small number of patients experienced mild to moderate side effects and serious adverse events.

Summary: This retrospective multicentre study aimed to investigate the characteristics and outcome of ANCA-associated vasculitis (AAV) induced by anti-thyroid drugs (ATD) compared to primary AAV. The results showed that ATD-induced AAV mainly presented as microscopic polyangiitis (MPA), with the most common manifestations being skin involvement, arthralgia, and glomerulonephritis. However, ATD-induced MPA was less severe and had a lower risk of relapse compared to primary MPA.

The study found that the DETAIL questionnaire is an effective tool for identifying missed cases of enteropathic arthritis in patients with IBD.

Summary: In this study, the HUVAC database was used to evaluate cases of pachymeningitis in patients with IgG4-related disease. It was found that most of these patients had normal serum IgG4 levels, therefore not useful for diagnosis. Typical radiology and tentorial thickening should raise suspicion of IgG4-related disease and prompt early biopsy. With steroids+ rituximab treatment, there was no long-term relapse related to meningeal involvement.

Summary: This study investigated the involvement of the JAK/STAT signaling pathway in the pathogenesis of inflammatory skin diseases (ISDs). The results showed that multiple components of the JAK/STAT pathway were overexpressed in the skin of ISDs patients. These findings suggest that targeting the JAK/STAT pathway could be a potential therapeutic strategy for treating ISDs.

Summary: This study aimed to investigate the effects of immunosuppressive doses on the results of the QuantiFERON-TB Gold Plus (QFT-Plus) test in patients with Rheumatoid Arthritis (RA), as well as the impact of the TB2 tube in the QFT-Plus test. The results showed that higher doses of immunosuppressive drugs in RA patients led to a lower positive rate in the QFT-Plus test, and the addition of the TB2 tube improved the test sensitivity.

Summary: In routine rheumatology practice, it was observed that a significant number of male ankylosing spondylitis (AS) patients did not experience inflammatory back pain (IBP). This study investigated the prevalence of IBP in male AS patients and compared it to that in female patients. The results showed that the prevalence of IBP was higher in female AS patients (87.7%) compared to male patients (66.1%). Additionally, female AS patients had higher disease activity levels than males.
