Summary: The study aimed to assess the diagnostic accuracy of classifying IDH and 1p19q status using MR imaging radiomics. It was found that combining conventional radiomics with convolutional neural network-derived features best classified IDH status, while texture-based radiomics best classified 1p19q status. Radiogenomics may serve as a potential alternative to invasive biopsy techniques for determining IDH and 1p19q status in lower-grade gliomas but requires further translational research for clinical uptake.

Summary: Treatment recommendations for grade 3 gliomas depend on their histopathologic and molecular phenotype. The current treatments involve surgery, radiation therapy, and chemotherapy. The addition of temozolomide after radiation therapy improves outcomes for non-codeleted grade 3 IDH mutant astrocytoma. For codeleted oligodendroglioma, the recommended treatment includes procarbazine, CCNU, and vincristine regimen. Novel agents targeting IDH mutation and its metabolic effects are being investigated to further improve patient outcomes.

Summary: The study aimed to describe the long-term course of oligodendroglioma disease in a population-based setting and determine factors affecting survival. Patients with WHO-grade 2 oligodendrogliomas and known 1p19q-codeletion were analyzed, with a median follow-up of 12.0 years and a median survival of 17.8 years. Factors associated with shorter survival include age, tumor diameter, and poor preoperative functional status.

Summary: The study demonstrated the use of a nanopore-based copy-number variation sequencing (nCNV-seq) approach to simultaneously identify deletions of CDKN2A/B and 1p/19q in IDH-mutant gliomas. The nCNV-seq approach showed high concordance with other methods in detecting the whole-arm 1p/19q codeletion and the CDKN2A/B deletion in astrocytomas and oligodendrogliomas. Furthermore, nCNV-seq can differentiate between different types of deletions of CDKN2A/B. Overall, nCNV-seq holds promise as a new and cost-effective approach for the molecular characterization of IDH-mutant gliomas.

Summary: In this study, the transcriptomic and proteomic profiles of low-grade gliomas (LGG) were investigated using RNA-seq data and tandem mass-spectrometry analysis. The results showed that gene expression in LGG subtypes was mainly influenced by IDH mutations and 1p19q co-deletion. Proteomic analysis identified subtype-specific proteins that could be used to classify the LGG subtypes.
