At Physikalisch-Technische Bundesanstalt, the National Metrology Institute of Germany, a new type of deep ultraviolet scatterometer has been developed and set up. The concept of the system is very variable and versatile, so that many different types of measurements, e.g., classical scatterometry, ellipsometric scatterometry, polarization-dependent reflectometry, and ellipsometry can be performed. The main application is the characterization of linewidth/critical dimension (CD), grating period (pitch), and edge profile of periodically nanostructured surfaces mainly, but not only, on photomasks. Different operation wavelength between 840 and 193 nm can be used, giving also access to a variety of different at-wavelength metrology connected with state-of-the-art photolithography. It allows to adapt and to vary the measurand and measurement geometry to optimize the sensitivity and the unambiguity for the measurement problem. In this paper the concept, design, and performance of the system is described in detail. First measurement examples are shown and current and future applications are discussed.

Probiotic potential of autochthonous bacteria from tambaqui Colossoma macropomum

Suzana Kotzent, Silvia Umeda Gallani, Gustavo Moraes Ramos Valladao, Lindomar de Oliveira Alves, Fabiana Pilarski

Summary: This study identified and characterized six potential probiotic strains for inclusion in the diet of tambaqui Colossoma macropomum, including Enterococcus faecalis, Enterococcus hirae, Lactococcus lactis, Pediococcus pentosaceus, Staphylococcus hominis, and Staphylococcus saprophyticus. These probiotic strains showed resistance against bile salts and antagonistic activity against a fish pathogen, Lactococcus garvieae.

Patterns of the innate immune response in tambaqui Colossoma macropomum: Modulation of gene expression in haemorrhagic septicaemia caused by Aeromonas hydrophila

Silvia Umeda Gallani, Gustavo Moraes Ramos Valladao, Lindomar de Oliveira Alves, Suzana Kotzent, Diogo Teruo Hashimoto, Geert Wiegertjes, Karina Kirsten, Luiz Carlos Kreutz, Fabiana Pilarski

Summary: The production of tambaqui fish has reached a milestone in South American continental waters, but its immunity remains poorly understood. This study established patterns of innate immunity for the species through two experiments, showing up-regulation of immune genes in infected fish in early and chronic phases of infection, except for HSP70 which showed down-regulation.

Haematological, biochemical and immunological biomarkers, antibacterial activity, and survival in Nile tilapia Oreochromis niloticus after treatment using antimicrobial peptide LL-37 against Streptococcus agalactiae

Elielma Lima de Sousa, Inacio Mateus Assane, Norival Alves Santos-Filho, Eduardo Maffud Cilli, Raphael Barbetta de Jesus, Fabiana Pilarski

Summary: The study evaluated the therapeutic effectiveness and potential adverse effects of peptide LL-37 against streptococcosis caused by Streptococcus agalactiae in Nile tilapia. The results showed that LL-37 had weak hemolytic activity against red blood cells of Nile tilapia and strong in vitro antimicrobial activity against S. agalactiae, but it was found to be ineffective in controlling streptococcosis in the fish.

Effects of triploid induction on innate immunity and hematology in Astyanax altiparanae

Nycolas Levy-Pereira, Mateus Maldonado Carriero, George Shigueki Yasui, Caroline Munhoz Meira, Ricardo Luiz Moro de Sousa, Antonio Augusto Mendes Maia, Jose Augusto Senhorini, Fabiana Pilarski

Summary: Triplod induction in fish can lead to changes in hematological parameters, erythrocyte morphology, and immune responses. Fish with triploid induction showed lower cell counts and altered cellular abnormalities, indicating potential immune impairments and lower resistance to diseases and poor environments.

Cytotoxicity and antimicrobial activity of synthetic peptides alone or in combination with conventional antimicrobials against fish pathogenic bacteria

I. M. Assane, N. A. Santos-Filho, E. L. de Sousa, M. C. O. deArruda Brasil, E. M. Cilli, F. Pilarski

Summary: Synthetic host defense peptides (HDPs) showed potent antimicrobial activity against various pathogenic bacteria in fish, making them a potential treatment option in aquaculture to reduce the use of antimicrobials. The effectiveness of HDPs such as KR-12-a5, LL-37, (p-BthTX-I)(2), and Hylin a1 can be further tested alone or in combination with conventional antimicrobials to explore alternative treatment strategies in aquaculture. The study highlights the synergistic effects of certain HDPs with traditional antimicrobials, suggesting a promising approach to combat bacterial diseases in aquatic animals.

Inacio Mateus Assane, Elielma Lima de Sousa, Gustavo Moraes Ramos Vallada, Geovana Dotta Tamashiro, Eduardo Criscoulo-Urbinati, Diogo Teruo Hashimoto, Fabiana Pilarski

Summary: This study investigated the characteristics of A. jandaei strains associated with massive mortalities in freshwater fish in Brazil, including morphology, biochemistry, molecular features, virulence, resistance, and pathogenicity. Variations were observed in virulence genes, experimental infections, and antimicrobial susceptibility among different strains.

Enterogyrus spp. (Monogenea: Ancyrocephalinae) and Aeromonas jandaei co-infection associated with high mortality following transport stress in cultured Nile tilapia

Inacio Mateus Assane, Karen Dayana Prada-Mejia, Silvia Umeda Gallani, Natasha Fernandes Weiser, Gustavo Moraes Ramos Valladao, Fabiana Pilarski

Summary: In Nile tilapia, co-infection by Monogenean stomach parasites (E. coronatus, E. foratus, and E. malbergi) and A. jandaei following transport stress can lead to high mortalities. The study provides insights into the rare occurrence of internal monogenean infections and highlights the potential synergistic effects of parasitic and bacterial infections.

The association of modified QuEChERS and DLLME to offer high analytical detectability to assess residual depletion profile of erythromycin in fish

Sarah Chagas Campanharo, Agnaldo Fernando Baldo da Silva, Anais Bleuzen, Jonas Joaquim Mangabeira da Silva, Lucas Victor Pereira de Freitas, Inacio Mateus Assane, Fabiana Pilarski, Jonas Augusto Rizzato Paschoal

Summary: An analytical method using modified QuEChERS procedure and DLLME technique was developed, optimized, and validated for the determination of erythromycin A (ERY) residues in fish fillet. The method exhibited LOD and LOQ of 0.1 μg/kg and 1 μg/kg, respectively. It demonstrated linearity in the range of 1 to 20 μg/kg, with precision (CV < 6.3%) and accuracy (recovery ranging from 103 to 110%). The method was further applied to study the residual depletion profile of ERY in fish (Piaractus mesopotamicus) after oral administration, with a withdrawal time estimated at 240 degree-days for concentrations below the maximum residue limit (MRL 100 μg/kg).
