Intrafollicular soluble receptor for advanced glycation end products (sRAGE) and embryo quality in assisted reproduction

The developmental potential of human embryos has important implications in assisted reproduction and depends, among other factors, on oocyte competency. The receptor for advanced glycation end products (RAGE) is a member of the superfamily of immunoglobulin cell-surface molecules that are constitutively expressed during embryonic development. RAGE is down-regulated in homeostasis in adult life. This study measured the concentration of soluble RAGE (sRAGE) in follicular fluid obtained from the leading follicle after ovarian stimulation of 54 women undergoing intracytoplasmic sperm injection. Corresponding embryos and sRAGE concentrations in follicular fluid were evaluated and correlations were investigated by multi-adjusted regression analysis. High intrafollicular sRAGE concentrations predicted poor-quality embryos (n = 45, OR = 0.986; P = 0.026), adjusted for patient age, body mass index and oocyte quality, showing an inverse association between intrafollicular sRAGE concentrations and embryo development. Summary: A novel vaccine based on a chimeric human adenovirus 5 vector was developed, which induced potent humoral and T-cell immunity lasting at least 5 months by enhancing Spike expression, infection of muscle and dendritic cells, and utilizing stabilized Spike in a prefusion conformation. Vaccinated mice showed neutralizing antibodies against various Spike variants and maintained stable immunity for over 5 months, suggesting potential for clinical trials.

Endometrial protein expression and phosphorylation landscape decipher aberrant insulin and mTOR signalling in patients with recurrent pregnancy loss

Summary: This study investigates the proteomic and phosphoproteomic differences in the endometrium of women with recurrent pregnancy loss (RPL) compared to healthy control women during different phases of the menstrual cycle. The results identify differentially expressed proteins and phosphorylated proteins, and highlight the insulin/cyclic nucleotide signalling pathway and AMPK/mTOR signalling pathway as major contributors to the abnormality of RPL endometrium. The findings provide insights into potential proteins associated with the pathogenesis of RPL and contribute to the identification of potential targets for RPL treatment.

Random-start ovarian stimulation in an oocyte donation programme: a large, single-centre, experience
