Preantral follicles are the majority of the ovarian follicle population and their use as a source of homogeneous oocytes for bovine reproductive biotechnologies could result in a substantial advance in this field. However, while in other species embryos and offspring have been produced, in bovine species the results have been limited to the follicular activation of small (primordial) preantral follicles and formation of early antral follicles from large (secondary) preantral follicles after in vitro culture. Therefore, this review will highlight the basic aspects of bovine folliculogenesis by focusing on preantral follicles, the methods of harvesting preantral follicles, the main results from in vitro follicular culture during the last 20 years, and the potential candidate substances (basic supplements, growth factors, and hormones) for improving the efficiency of in vitro follicle

Interference of fixatives and fixation period on the morphologic analysis of ovarian preantral follicles

Summary: The type and period of fixation affected the percentage of morphologically normal ovarian follicles. Paraformaldehyde was not recommended as a fixative, while formalin was suitable for 12-hour fixation and Carnoy's solution was efficient for 24-hour fixation. Davidson's solution was the most suitable fixative for evaluating the morphology of ovarian preantral follicles, regardless of the fixation period.

Summary: This study evaluated the effects of whole flaxseed as a feed supplementation on the in vitro development of caprine early antral follicles and embryo production. The results showed that flaxseed did not affect follicular growth and diameter, but increased the recovery rate of oocytes and negatively affected sperm penetration rate.

Summary: This study aimed to investigate the relative abundances of ABC transporters mRNA transcripts in ovine ovarian tissue after vitrification and in vitro culture. The results showed a negative correlation between degenerated follicles and mRNA transcript abundances of ABCB1 and ABCG2, with a significant decrease in the percentage of growing follicles after in vitro culture. Further studies are needed to explore the functions of ABC transporters in ovine folliculogenesis after in vitro culture.

Summary: This study evaluated the effects of zearalenone and matairesinol on the morphology of in vitro-cultured ovarian follicles. Zearalenone impaired the follicle morphology, while the combination of matairesinol and zearalenone counteracted the negative effects. However, matairesinol did not promote follicle development in vitro.

Summary: This study evaluated the effects of different concentrations of eugenol, ascorbic acid, and anethole in goat ovarian tissue. The results showed that eugenol promoted follicle development and survival, and reduced oxidative stress. Furthermore, eugenol decreased gene methylation and endoplasmic reticulum stress.

How in vitro maturation changes the proteome of ovine cumulus-oocyte complexes?

Summary: The present study evaluated the effects of in vitro maturation (IVM) on the proteome of cumulus-oocyte complexes (COCs) from ewes. The results showed that IVM caused upregulation and downregulation of proteins in COCs. Upregulated proteins in mature COCs after IVM were related to immune response, complement cascade, cell cycle, and extracellular matrix organization, while downregulated proteins in immature COCs after IVM were linked to metabolic processes, immune response, and complement cascade.

Characterization of preantral follicle clustering and neighborhood patterns in the equine ovary

Summary: This study investigated the clustering and neighborhood patterns of preantral follicles in the horse ovary, showing that younger mares have more clustering and neighbors compared to older mares. The clustering was found in all areas of the ovary, and follicles with neighbors were more likely to be morphologically normal.

Oocyte in vitro maturation with eugenol improves the medium antioxidant capacity and total cell number per blastocyst

Summary: This study investigates the effects of eugenol supplementation on bovine oocyte in vitro maturation and the subsequent in vitro embryo production and quality through IVF. The results show that adding 40μM eugenol to the maturation medium improves antioxidant capacity and cleavage rate. Additionally, lower calreticulin levels and higher total cell numbers in expanded blastocysts were observed in the lower eugenol concentration treatments. Based on these findings, enriching the maturation medium with 10μM eugenol is recommended for in vitro bovine embryo production.

Comprehensive proteomic profiling of early antral follicles from sheep

Summary: The study evaluated the proteome of early antral follicles from Ovis aries using LC-MS/MS analysis. A total of 2503 follicle proteins were identified, including abundant proteins such as vimentin, actin, lamin, heat shock proteins, and histones. miRNAs were found to modulate genes involved in cell cycle, cell differentiation, aging, apoptosis, adipocyte differentiation, and cell division in sheep follicles. This comprehensive atlas of proteins expressed in sheep early antral follicles will contribute to the identification of biomarkers for follicular development and oocyte maturation.

Summary: This study found that adding 100μM or 150μM ALA to the vitrification solution can protect sheep ovarian tissue, maintain follicle morphology, and promote follicular development after in vitro culture or autotransplantation. However, high levels of ALA may cause oxidative stress and tissue fibrosis after transplantation.

Summary: This study investigated the effects of zinc oxide nanoparticles functionalized with curcumin (ZnO(np) + CUR) on bovine in vitro embryo production and cellular antioxidant response during in vitro embryo culture (IVC). Cumulus-oocyte complexes (COCs) were matured, fertilized, and cultured in the presence of different concentrations of ZnO(np) + CUR. The results showed that the addition of 12μM ZnO(np) + CUR during IVC reduced intracellular ROS production and blastocyst production rate.
