Melatonin is a ubiquitous molecule, present in a wide range of organisms, and involved in multiple functions. Melatonin relays the information about the photoperiod to the tissues that express melatonin-binding sites in both central and peripheral nervous systems. This hormone has a complex mechanism of action. It can cross the cell plasma membrane and exert its actions in all cells of the body. Certain melatonin actions are mediated by receptors that belong to the superfamily of G-protein-coupled receptors (GPCRs), the MT1 and MT2 membrane. Melatonin can also bind to calmodulin as well as to nuclear receptors of the retinoic acid receptor family, RORa1, RORa2 and RZRb. The purpose of this review is to report on recent developments in the physiological role of melatonin and its receptors. Specific issues concerning the biological function of melatonin in mammalian seasonal reproduction and spermatozoa are considered. The significance of the continuous presence of melatonin in seminal plasma with a fairly constant concentration is also discussed.

Involvement of progesterone and estrogen receptors in the ram sperm acrosome reaction

Summary: The steroid hormones E2 and P4 can induce the acrosome reaction in ram spermatozoa through receptor-mediated mechanisms. Estrogen receptors (ER) and progesterone receptors (PR) are present in these cells, suggesting a receptor-mediated effect of the hormones on sperm function.

Summary: This study demonstrates variations in melatonin concentration in seminal plasma between rainy and dry seasons in three Colombian sheep breeds, and investigates how melatonin modulates sperm capacitation in equatorial-located breeds. The addition of melatonin to the cocktail medium is shown to partially prevent the increase in capacitated spermatozoa across breeds and seasons, shedding light on the direct effects of melatonin on spermatozoa in ovine breeds under equatorial photoperiod conditions.

Testicular Ultrasound Analysis as a Predictive Tool of Ram Sperm Quality

Summary: Testicular ultrasound can be a valuable tool for predicting ram seminal quality, with white and grey pixels correlating with poor sperm parameters and tubular density and lumen area correlating with good sperm quality. Ultrasound-video analysis allows for both macroscopic and microscopic evaluation of testicular echotexture, providing a non-invasive method for assessing ram fertility.

Effects of fetal bovine serum on trophectoderm and primitive endoderm cell allocation of in vitro-produced bovine embryos

Summary: This study found that supplementation of fetal bovine serum (FBS) in embryonic culture medium can affect cell differentiation during early bovine embryo development. Additionally, FBS supplementation alters the allocation of trophectoderm (TE) and primitive endoderm cells.

Effect of melatonin and nitric oxide on capacitation and apoptotic changes induced by epidermal growth factor in ram sperm

Summary: This study investigates the effect of melatonin and nitric oxide on capacitation and apoptotic-like changes in EGF-capacitated ram spermatozoa. The results indicate that NO levels and apoptotic-related markers increase after EGF incubation. Melatonin has a bimodal role in sperm EGF-capacitation, but neither of the two concentrations prevents the increase in apoptotic-like markers or NO levels. However, melatonin at 1μM prevents the activation of JNK.

Summary: The aim of this study was to evaluate the effect of a phytomelatonin-rich diet on ram sperm quality and seminal plasma composition. The results showed that the diet, including grape pulp, pomegranate and tomato pomaces, increased melatonin levels in seminal plasma, improved sperm viability and morphology, and protected sperm cells against oxidative damage.
