Chemical Composition of Lipids Present in Cat and Dog Oocyte by Matrix-Assisted Desorption Ionization Mass Spectrometry (MALDI- MS)

Contents The aim of the present study was to investigate the level of information on the chemical structures and relative abundances of lipids present in cat and dog oocytes by matrix-assisted laser desorption mass spectrometry (MALDI-MS). The MALDI-MS approach requires a simple analysis workflow (no lipid extraction) and few samples (two or three oocytes per analysis in this work) providing concomitant profiles of both intact phospholipids such as sphingomyelins (SM) and phosphatidylcholines (PC) as well as triacylglycerols (TAG). The lipids were detected in oocytes by MALDI using dihydroxybenzoic acid (DHB) as the matrix. The most abundant lipid present in the MS profiles of bitch and queen oocytes was a PC containing 34 carbons and one unsaturation [PC (34:1)]. Oocytes of these two species are characterized by differences in PC and TAG profiles detected qualitatively as well as by means of principal component analysis (PCA). Cat oocytes were mainly discriminated by more intense C52 and C54 TAG species and a higher number of unsaturations, indicating predominantly linoleic and oleic fatty acyl residues. Comparison of the lipid profile of bitch and queen oocytes with that of bovine oocytes revealed some similarities and also some species specificity: TAG species present in bovine oocytes were also present in bitches and queens; however, a more pronounced contribution of palmitic, stearic and oleic fatty acid residues was noticed in the lipid profile of bovine oocytes. MALDI-MS provides novel information on chemical lipid composition in canine and feline oocytes, offering a suitable tool to concomitantly monitor, in a nearly direct and simple fashion the composition of phospholipids and TAG. This detailed information is highly needed to the development of improved protocols for in vitro culture and cryopreservation of cat and dog oocytes.

Nutritional labelling of food products purchased from online retail outlets: screening of compliance with European Union tolerance limits by near infrared spectroscopy

Summary: Nutritional information on food labels can influence consumers' dietary choices. This study used near infrared spectroscopy to verify the accuracy of nutritional labels on 103 food products. The results showed that 35% of processed food products did not have nutritional labels, and some of the labeled values did not match the experimental data. Therefore, this study demonstrated the capability of near infrared spectroscopy in monitoring compliance with EU regulations and providing reference methods for further confirmation.

Summary: This study developed and validated a method to automatically serotype Legionella pneumophila and distinguish it from Legionella non-pneumophila using Fourier-transform infrared spectroscopy (FT-IR) and machine learning. Data validation strategies were employed to ensure consistent performance of the method across a large microbial population. The method achieved high accuracy on different test sets from European collections.

Seasonal Variation in Raw Milk VOC Profile within Intensive Feeding Systems

Summary: The study aimed to assess the seasonal variation in VOCs in raw milk from three indoor feeding systems. VOC profiles were determined by GC after HS-SPME. The results showed that the feeding system did not affect VOCs, but the season had a significant impact. The outcomes suggested that VOCs could be used to trace raw milk for further mild processing.

Metabolic signature of Mycobacterium avium subsp. paratuberculosis infected and infectious dairy cattle by integrating nuclear magnetic resonance analysis and blood indices

Summary: This study investigated the metabolic changes associated with natural MAP infection in dairy cattle using NMR spectroscopy and routine blood chemistry. The results showed altered metabolic pathways, including increased tyrosine metabolism and enhanced phenylalanine, tyrosine, and tryptophan biosynthesis in MAP-infected cattle. Increased synthesis and degradation of ketone bodies were observed in both infected and infectious cattle. The fusion of multiple data sources proved to be useful in exploring these metabolic pathway changes and potentially diagnosing negative animals within paratuberculosis-infected herds.

Microbiological and chemical safety of Bombyx mori farmed in north-eastern Italy as a novel food source

Summary: Silk farming in Italy has declined in the past due to competition with Asian markets and pesticide use, but it has potential for future development in the food sector. This study aimed to assess the nutritional composition and presence of hazards in silkworms. The study found that some samples did not meet the microbiological criteria for certain bacteria and fungi. Additionally, traces of fungicides and heavy metals were observed, and mycotoxins were detected in certain products. Overall, this study highlights the potential impact of environmental contamination on the safety of silkworms and their derivative products.

Microbial and chemical stability of Acheta domesticus powder during one year storage period at room temperature

Summary: This study evaluated the microbiological and chemical stability of A. domesticus powders during one year of storage at room temperature. The results showed that the cricket powders had suitable microbiological stability, but exhibited a clear tendency toward rancidity in terms of chemical stability. Samples dried at 120°C and lyophilized never exceeded the limit set for B. cereus count in A. domesticus powder. Lyophilized samples also had the lowest levels of aldehydes responsible for rancid smell.

Summary: In this study, ambient mass spectrometry (AMS) screening method was applied to analyze a suspected animal poisoning case. The results showed evidence of vegetable material intoxication and confirmed the presence of toxic substances using other analysis methods. This study demonstrates the potential of AMS to offer a rapid and efficient alternative for analyzing toxic substances in autopsy specimens from animals.

In vivo embryo development in bitches inseminated laparoscopically after ovulation time estimated based on a single progesterone determination

Summary: The aim of this study was to evaluate early embryonic development in bitches using sequential vaginal cytologies, single progesterone measurement, and single laparoscopic insemination. The results showed that the optimal insemination time can be accurately predicted 48 hours post-ovulation based on a single progesterone measurement, and using high-quality semen results in a higher number of embryos. This finding is important for dog owners facing logistical or economic limitations in accurately planning the breeding time.
