Influence of Season on the Freezability of Free-Range Poultry Semen

Contents The purpose of the present study was to examine the seasonal variation in freezing damage in free-range rooster sperm. Over a period of 1 year, heterospermic semen samples were collected weekly by massage from the roosters of 14 Spanish chicken breeds, all housed under natural photoperiod and climatic conditions. All samples were frozen in straws using DMA as a cryoprotectant, placing them first in nitrogen vapour and then plunging them into liquid nitrogen. No seasonal effects on fresh sperm quality were found. Neither did season affect the percentage of viable frozenthawed spermatozoa nor the percentage with an intact acrosome. However, the collection season influenced (p < 0.05) most frozenthawed sperm motility values. The percentage of immotile frozenthawed spermatozoa was lower (p < 0.05) in spring-collected sperm than in summer- or autumn-collected samples. The percentage of spermatozoa showing progressive motility was higher in spring-collected sperm compared with winter-, summer- or autumn-collected samples (p < 0.05). The curvilinear velocity (VCL), straight-line velocity (VSL) and average path velocity (VAP) values of spring-collected sperm were also higher (p < 0.05). In conclusion, spring would appear to be the best season for collecting and freezing the semen of free-range Mediterranean chicken breeds.

Summary: This study compared the effects of two cryoprotectants, DMA and DMSO, on the frozen-thawed sperm characteristics of black-footed penguins and gentoo penguins. The results showed that 8% DMSO was more effective for black-footed penguin sperm, while there were no significant differences in gentoo penguin samples. Additionally, black-footed penguin sperm was more sensitive to the freezing-thawing process compared to gentoo penguin sperm.

Variation of existence and location of aquaporin 3 in relation to cryoresistance of ram spermatozoa

Summary: The study aimed to investigate the influence of osmotic changes on the location of aquaporins (AQPs) in spermatozoa during freeze-thawing process. Individual variability in terms of freezeability was observed among rams and their ejaculates. AQP3 expression in the mid- and principal piece of spermatozoa was found to be higher in frozen-thawed samples compared to fresh specimens in the good freezability group, but such differences were not detected in the poor freezability group. AQP3 expression is associated with sperm cryotolerance and can be used as a biomarker.

Review: Sperm cryopreservation in wild small ruminants: morphometric, endocrine and molecular basis of cryoresistance

Summary: Reproductive technologies can help protect wild ruminant species from extinction and increase the number of animals. Efforts have focused on preserving sperm, oocytes, and embryos. Cryopreservation of sperm is the most commonly used and cheapest method for germplasm storage. New reproductive biotechnologies, such as ultra-rapid freezing techniques, have been developed. However, the success of artificial insemination using frozen-thawed sperm is not always satisfactory in wild small ruminants. Epididymal sperm shows higher cryoresistance than ejaculated sperm, and the seasonal variation in testosterone and prolactin levels is involved in sperm cryoresistance.

Summary: This study examines the effect of equilibration time with extender on the variables of ultra-rapidly frozen-thawed sperm from wild ruminants. The results show that equilibration improves the quality and viability of sperm. Epididymal sperm demonstrates greater cryoresistance, while ejaculated sperm shows no dimensional changes before and after equilibration.

The results showed that the transit though the vas deferens resulted in a higher percentage of compacted chromatin spermatozoa in the ejaculate.
