Regulation of luteal function and corpus luteum regression in cows: Hormonal control, immune mechanisms and intercellular communication

The main function of the corpus luteum (CL) is production of progesterone (P4). Adequate luteal function to secrete P4 is crucial for determining the physiological duration of the oestrous cycle and for achieving a successful pregnancy. The bovine CL grows very fast and regresses within a few days at luteolysis. Mechanisms controlling development and secretory function of the bovine CL may involve many factors that are produced both within and outside the CL. Some of these regulators seem to be prostaglandins (PGs), oxytocin, growth and adrenergic factors. Moreover, there is evidence that P4 acts within the CL as an autocrine or paracrine regulator. Each of these factors may act on the CL independently or may modify the actions of others. Although uterine PGF(2 alpha) is known to be a principal luteolytic factor, its direct action on the CL is mediated by local factors: cytokines, endothelin-1, nitric oxide. The changes in ovarian blood flow have also been suggested to have some role in regulation of CL development, maintenance and regression.

Summary: The development of equine endometrosis can be slowed down by inhibiting the pro-fibrotic effects of myeloperoxidase using the inhibitor ABAH. Metallopeptidase-2 might be involved in an acute response to myeloperoxidase in the mid-luteal phase, while MMP-9 might be implicated in a prolonged exposition to myeloperoxidase in the follicular phase.

Summary: This study investigates newly discovered mechanisms of atresia and cell death processes in bovine ovarian follicles, finding that necroptosis is an alternative mechanism responsible for the death of bovine granulosa and theca cells. Analysis of mRNA expression of RIPK1 and RIPK3 supports this conclusion, highlighting the importance of both apoptosis and necroptosis in follicular cell death.

The Effects of Prostaglandin E2 Treatment on the Secretory Function of Mare Corpus Luteum Depends on the Site of Application: An in vivo Study

Summary: In this study, we investigated the impact of prostaglandin E-2 (PGE-2) on the secretory function of equine corpus luteum (CL), comparing the effects of intra-CL injection and intrauterine administration. We also compared the effects of these administration methods on CL function using both PGE-2 and human chorionic gonadotropin (hCG) as luteotropic factors. The results demonstrated that PGE-2 application, regardless of the site of administration, supported equine CL function and led to differences in progesterone and PGE-2 concentrations in the blood plasma.

Peroxisome proliferator-activated receptor expression in the canine endometrium with cystic endometrial hyperplasia-pyometra complex

Summary: This study aimed to evaluate and compare the mRNA expression and immunolocalization of PPARα, PPARδ, and PPARγ in the canine endometrium. The results showed that the expression levels of PPARs changed during cystic endometrial hyperplasia (CEH) and CEH-pyometra, suggesting their potential involvement in hormonal disturbances and inflammatory processes.

Evolution of the Concepts of Endometrosis, Post Breeding Endometritis, and Susceptibility of Mares

Summary: Our understanding of post breeding endometritis and susceptibility of mares has changed over the last 100 years. Initially, it was believed that bacterial infection in the uterus after mating was the cause, but it was later discovered that sperm induce neutrophilia, indicating that post breeding endometritis is a physiological defense mechanism. In susceptible mares, this defense mechanism can be prolonged, leading to chronic endometritis. This paper reviews the pathogenesis of endometritis and endometrosis.

Molecular Mechanism of Equine Endometrosis: The NF-κB-Dependent Pathway Underlies the Ovarian Steroid Receptors' Dysfunction

Summary: Endometrosis, a common disease that affects mares' fertility, is associated with alterations in epithelial and stromal cells, as well as degeneration and fibrosis of the endometrium. The study reveals a potential involvement of the NF-kappa B pathway in the pathogenesis of endometrosis, showing differential transcription of profibrotic genes between different phases of the estrous cycle. Additionally, the transcription of estrogen and progesterone receptors is found to be decreased with the severity of fibrosis and degeneration of the endometrium in horses.

Collagen Type III as a Possible Blood Biomarker of Fibrosis in Equine Endometrium

Summary: This study aimed to identify possible blood biomarkers for endometrosis diagnosis and fertility assessment on mares. The results suggest that serum COL3 concentration might be considered as a potential aid for the diagnosis of endometrosis and fertility prognosis in the mare. In contrast, COL1 and hydroxyproline did not prove to be effective as biomarkers of endometrial fibrosis in this species.

The Role of Peroxisome Proliferator-Activated Receptors in PGF2α-Induced Luteolysis in the Bovine Corpus Luteum

Summary: The aim of this study was to determine the expression of PPAR isoforms in the bovine corpus luteum throughout the estrous cycle, and their involvement in PGF(2 alpha)-induced processes related to luteolysis. The results showed that the mRNA and protein expression of PPARs changes in the bovine corpus luteum throughout the estrous cycle and under the influence of PGF(2 alpha). PPAR gamma, among all examined isoforms, could be involved in the regulation of PGF(2 alpha)-induced luteolysis in cattle.

Inhibition of Myeloperoxidase Pro-Fibrotic Effect by Noscapine in Equine Endometrium

Summary: This study aims to evaluate the inhibitory effect of noscapine on collagen type 1 (COL1) induced by myeloperoxidase. The results show that myeloperoxidase increases the transcription of COL1A2 gene and the abundance of COL1 protein, while noscapine is able to reduce this effect, with a time/estrous cycle phase-dependent manner (in follicular phase explants, at 24 h of treatment).

Summary: This study aims to explore the activation of AMP-activated protein kinase (AMPK) in boar spermatozoa under oxidative stress. The results show that oxidative stress promotes the activation of AMPK, which is positively correlated with sperm quality and functionality. This sheds light on the mechanism of sperm resistance to oxidative stress and provides valuable information for selecting antioxidants to improve reproductive outcomes.
