Changes in the expression of interleukin-1β and lipopolysaccharide-induced TNF factor in the oviduct of laying hens in response to artificial insemination

The aim of this study was to determine the physiological significance of interleukin-1 beta (IL1 beta) and lipopolysaccharide-induced TNF factor (LITAF) in the fate of sperm in the oviduct of laying hens after artificial insemination (Al). Laying hens were inseminated with fresh semen, PBS or seminal plasma and tissues from different oviductal segments were collected to observe the general histology, changes in the mRNA expression of IL1 Band LITAF and the localization of positive cells expressing immunoreactive IL1 B (irIL1 B). Semi-quantitative RT-PCR was used to observe the changes in mRNA expression of these molecules in the infundibulum, uterus, utero-vaginal junction (UVJ), and vagina after insemination. Intact sperm in the lumen and between the primary or secondary folds of the vagina were found until 6 h after insemination but were degraded at 12 h. The mRNA expression of IL1B and LITAF was significantly increased in the vagina until 6 h after Al but remained unchanged in the other oviductal segments. In the tissue of the vagina and UVJ, irIL1 B was localized in the mucosal stroma. The number of irIL1B-positive cells was increased in the vagina but almost unchanged in UVJ after insemination with semen. Significant changes were not observed in the mRNA expression and irIL1 B-positive cells in the vagina after PBS or seminal plasma insemination. The increase of IL1B and LITAF in the vagina may lead to sperm degradation and elimination by cilia of surface epithelium, whereas their lower levels in UVJ may permit sperm to survive in sperm storage tubules.

Summary: The oral administration of a lower dose of DSS caused slight disruptions in the intestinal environment, resulting in poor intestinal morphology and decreased cecal microbiome diversity. This led to a decrease in egg yolk size but did not reduce the supply of VLDLy from the liver. The reduction in egg yolk size was likely due to dysfunction in the uptake of egg-yolk precursors in ovarian follicles.

Summary: This study aimed to investigate the response of the innate immune system in the proventriculus of broiler chicks to lipopolysaccharide (LPS) and how this response is influenced by Newcastle disease and infectious bronchitis (ND/IB) vaccination. The results showed that LPS derived from gram-negative bacteria can upregulate the innate immune system in the proventriculus, including the synthesis of antimicrobial peptides. ND/IB vaccination may not significantly affect antimicrobial peptide synthesis in response to LPS, but it can upregulate Toll-like receptor 21 expression.

Summary: This study aimed to investigate the effects of vaginal administration of Lactobacillus johnsonii on the mucosal barrier function of the oviduct. The results showed that vaginal administration of L. johnsonii improved the oviductal mucosa microbiota, enhanced the mucosal barrier function, and increased the ratio of beneficial bacteria while decreasing the ratio of pathogenic bacteria.

Summary: This study aimed to identify the cause for inferior quality egg production of aged hens by investigating the aging-associated molecular changes related to eggshell formation and determining the role of nitric oxide in decreasing egg quality. The results showed that molecular changes in the uterine mucosa affect eggshell membrane formation and eggshell biomineralization, while nitric oxide overproduction may play a role in this dysfunction.
